    Hello Madam,
    That was a great explanation. Wonder why UA-cam didn't recommend your channel all these days. Keep these kind of videos coming. People are still hesitating to get vaccinated, this video will clearly let them know it's impact.
    I have a couple of questions.
    1. These vaccines (all vaccines as of today) are built basing on the research they have done on the covid 19 virus and have created the vaccines basing on the results which were carried from the analysis of Covid 19.
    But as we don't know the origin of this virus we are not able to create the proper vaccine/drug which will give a 100 percent confidence that if a vaccine/drug is taken then covid will die inside the body. Am i right?
    2. I am a software engineer who knows a bit of machine learning. After the first dose/second dose gets injected into the person basing on the graph you have shown it shows a log normal distribution (towards the right side in the graph it keeps going and never ends). So if the scientists were correct then the antibodies will keep developing according to that graph. Am i right?.
    3. Does this work against all mutations which evolved and which may evolve later??

      According to me these are the explanations for your questions
      1. No we need not know about the origin of virus for development of the vaccine. We know its whole genetic material and the way it acts on body, that’s sufficient to create the vaccine. However, the desperate times required that we develop the vaccine fast ( normally this process takes years)...so the need of vaccine led to the vaccines of even lower efficacy being approved...details in my upcoming video

      Q2. Yes, there are memory cells...and they will keep producing antibodies...however without the stimulus, the concentration of antibodies goes down because of routine protein turnover in body. However when the body will be exposed to the virus, memory cells will produce antibodies
      P.S ...you are absolutely right about log distribution...many functions in body follow that pattern

      3. This question is tricky...nobody can predict that...however data suggests vaccine efficacy is much lower to some of the mutants...if virus mutates (like HIV) , it’s difficult to develop avaccine against such virus...in covid case...vaccines do show some efficacy...so something is better than nothing

      @@PhysiologyOpen So these memory cells recognise the pattern and they produce anti bodies and never stop producing them. If we keep having protein rich good then will this count keep ticking?. Or my perception is wrong - only when virus comes in these memory cells react quickly and produce antivirus cells and they go and attack the virus cells?.

      @@PhysiologyOpen There was a news which I saw yesterday a women with HIV had 32 mutations inside her. Wonder this wave 2 came out from such kind of situations. Also say if a person has sugar problem or HIV or any problem and gets covid, their body will be in a position to generate new mutations inside their body given that they are already less immune?.

    Nicely explained.... But if the antibody lasts for only 1month or more then how many booster doses required? & is the genome @ mRNA of adeno viruses are same as different varients of corona virus🦠😷?
    According to your explanation after the first doses the antibodies rises up to peak level in 1/2months then what is justification for making the gap of 84days....& after the second dose how long the antibody titre will remain high?

      2 months is what generally happens, but the response may vary from antigen to antigen...the gap also decided by vaccine availability, vaccine types.. booster doses will definitely be required but again ‘when’ depends on the vaccine

    B cells themselves act as APCs ie antigen presenting cells which are then read by helper T cells. There's clonal selection ie multiplication of T and B cells specific to the antigen ie spike protein. Antibodies will destroy the spike proteins in the primary immune response. There may be some antibodies left. At the end of primary immune response memory T and B cells are produced. So formation of memory cells takes more time with the primary immune response. The same gets accelerated with the 2nd dose of vaccine. Also the main antibody in primary immune response is IgM & it's IgG in secondary immune response ie 2nd dose of vaccine. IgG has more affinity than IgM and is a better quality antibody than IgM. This is apart from the quantum of IgG and IgM antibodies produced in 1st and 2nd doses.
    Adenovirus used has the part of its genome that allows it to replicate, removed making it non replicating

    thank you for explaining in such details!!( I just got vaccinated by covishield been 12 days am doing perfectly fine) but !! I have a query!! that if spike proteins are produced by our own cells post delivery of spike specific genes doesn't our own immune system attack our own cells and harm us!! 2: Are spike only produced at the site of injection and no where else in the body...i mean i know that 0.5ml of vac is a minute amount to get all across the body but even then isnt there chances of vac getting absorbed and getting transmitted to heart or lungs etc and damaging those areas post spike protein expression!! just curious!!

      If virus doesn’t multiply...protein itself doesn’t do damage...this protein helps virus move into cells and multiply. In vaccine virus is not there, only spike protein

    Mam coronavirus already has RNA genome. My question is when it is integrates with adenovirus vector, whether reverse transcription is done to convert in to DNA? This question arise because u said after entering immune cells, through transcription process DNA is transcripted to mRNA.
    My another question. is there any particular area of our body where all these immune responses occur? Is there connection between our body pain , headache and this?

      In an ideal world of healthy immune systems this vaccine could be best but science is aware that many humans do activities that weakens the immune system therefore there will be consequences?
      Theories dont come from ignorance but some vaccines have been harmful in the past and people become skeptical?
      Now, why variants are happening in population who are 80% vaccinated?
      Is this virus a product of errors in a lab which created a multi variant virus and somehow escaped?
      How about the WHO agency closing all labs that are not transparent on experiments or that are manipulating viruses ?

If the effect of the whole mechanism starts from only after 1 st week, then why its said that its common to expect people to fall bit sick like fever, body pain , head ache..etc as soon we take vaccine may be in the first week usually..How cautitious one should be during the 'after vaccination period' before the second dose as well after the second dose..Awaiting kind response

      You have to be as cautious as before vaccination...there is something known as viral load I.e the amount of virus we are exposed to...if that is too much..vaccine will have less effect...also mutant viruses are circulating, against which vaccines are less effective (though something is better than nothing)

      Chances of getting infected are there in between the two doses and even after two days. After two doses, however there are very less chances of severe disease and hospitalisation

      Cytokine storm occurs due to inappropriate response of body to kill the virus when it is unable to win over it. Vaccine doesn’t has virus. Moreover it prepares the body for an adequate response when it actually meets the virus

      Actually no...it does not suppress immune system...on the contrary...it is excessive immune reaction which is causing most damage

    As per new research the spike protein generated by covishield is breaking down and mixing with blood which is causing clot and cardiac arrest in few. Is astrazenika going to do anything on this?

      Yes there was some concern, and it was banned also in few countries. But then after looking at the data I think the ban was lifted.
      Secondly in case of any issue (for this data collection by government agencies and scientists), that data has to be shared with WHO and government which can/will put a stop.
      As far as company is concerned, if they change the formula depending on the data, they will have to do the research all over again.

      As per scientist a small tweak in the vaccine is sufficient to change its function but as you say I am not sure whether the trial has to go through again. Along with astrazenika even j&j are in touch with these German scientist for further changes.
      Since it's a very rare incident WHO and most of the top heads are silent on this.

      Some of my colleagues have also taken covaxin. I didn’t say it doesn’t work. Efficacy as shown in recent data is 77%

      It may be minimally effective but second dose is recommended. If it’s not available increasing the time duration from 8 to 12 weeks after the first dose is also ok...rather it’s recommended

      @@pawandixit5575 actually if you take the dose after 12 weeks , it’s much better as shown by antibody response...can’t say that about 16 weeks...I couldn’t find any scientific evidence for that

      No recommendation against alcohol as such but if you don’t eat properly then it may interfere with protein synthesis required for an adequate immune response

      For some vaccines , it has been found that alcohol interferes with response...for other vaccines it has no effect
      No recommendation for covid vaccines as such...
      Sleep good, eat good, no stress is what Research shows improves immune response
