Some steps in the tissue culture process include:
Explant excision
A disinfected tissue block is placed in a sterile petri dish, and superficial tissues are removed. A sharp scalpel is then used to excise the leaf primordia, and as much corm tissue as possible is removed.
Sterile solution
After excision, the explant is immediately transferred to a sterile solution of 50 mg/L cysteine to prevent blackening.
Incubation
The eggs plants are inoculated in a sterile environment and incubated at a temperature of 24 to 26 degrees C.
Light
The light intensity is initially maintained at 1500 to 3000 Lux, with a 16 hour light and 8 hour dark cycle. Once the plant has rooted, the light cycle is changed to 14 hours of light.
