I feel like it's probably technically possible to plan out an exact route and button sequence to beat the game then actually write songs and map those buttons to the notes so that you actually could beat it by just playing songs. But holy shit would that probably be stupid levels of complicated and time consuming.
Not really, just record the button presses and times they were pressed as a text file, and then write a script to translate each press into it's corresponding note and timing. You could then take that data and enter it into musescore or something and there you go. The longest part is that last step, but that step isn't even really necessary.
Techically all speedruners learn presses and times to run mario as fast as possible, and even use some bug/features to be even faster. Game also work using some frame rate for all things that happened in game. So i believe it is posible to do if you really wanna to do this, and if you can solve problems with input lags. And spend really big ammount of time to do this.

The cool thing about this gameplay is that the controls are not assigned to random notes, but to the notes of a major scale.
From what I saw when he was adding notes, you can program whatever frequency you want.
