With the Seek scene, I think the implication was that the “Seek juice” has some sort of hypnotic effect and Seek was coming to hypnotize a new set of Giggles into Screeches

What happened at the end is that Seek came back to the nest with Grumbles Mom infected, along with the sign she used to train Party Grumble. The grumble you saw in Mischievous Lights realm was Party grumble. Party Grumble would've joined his mom in Seeks possession if it wasn't for Guiding Light defeating Mischievous Light.

I never noticed the skibdi toilet dialogue also I always liked how the cartoon sounds were only with El Goblino and Bob really matches them as silly doors characters also if the queen does get voiced I think Unsurprise will voice her
