    Should we acquire spectra in 2x/5x/50x for best result?

      Hello, Thanks for your question. As is often the case it depends on the application and the sample!
      If you are running an automated measurement the 50x that will be used for the Ramana part of the analysis. In this case you select the objective most suited for the Morphological part of the analysis and the system will automatically switch to the 50x to acquire each spectrum. In our support materials there is guidance around which objective to select if you will be performing a full MDRS analysis depending on the particle size. If you are using the system in manual mode then it is possible to acquire a spectrum using other objectives, and which you choose will depend on the sample eg particle size and scattering efficiency. Please do reach out to your local Malvern Panalytical support for further guidance or training.

      We have separated particle which is very small in amount,we are getting spectra of backgroung not the single particle.any reason..

      There are various reasons that may lead to you not getting a good spectrum from your particle.
      It may be related to the particle size and/or the scattering efficiency of the material. Some materials are not Raman active. The spot size of the Raman beam is ~2um. If the particles of interest are smaller than this and/or are weak scatters then you can try extending the acquisition time. You can also consider applying a background correction.
