No.. they will cross only if channels are open. The method assumes that at rest no sodium, chloride channels are open. But logically you are right some ions do cross the membrane and hence it does not give perfect measurement of extra cellular fluid compartments. It is better represented by using the term ‘sodium space’: the body fluid compartment measured by radiolabelled sodium
After the injection of the indicator, it equilibrates with the various body fluids. If a sample of blood is taken after some time, the concentration of the indicator in that blood sample is C2
