- Modafinil is a narcolepsy drug and a very popular nootropic. I am going to show you how to synthesize it because it is an interesting reaction. Legality depends on where you live. I did my research and it is legal for me to make as long as it is neither consumed nor sold. Modafinil is also called Provigil. It can be made from benzhydrol, chloroacetamide, sodium hydroxide, thiourea, hydrogen peroxide and glacial acetic acid. Modafinil promotes wakefullness.
If you live in the USA it is NOT allowed to do this so do not repeat anything you see here no matter where you live. In Germany Modafinil was explicitly removed from the illegal narcotics list in 2008 so I can make it legally as long as other laws are being followed.

I was tired and seriously depressed all of the time before I tried this drug. I tried all sorts of things that didn't work, and I didn't think this would do anything for my depression, but it does. I'm not even sure if I qualify as depressed now. I have other issues, but this really helps. My doctors won't prescribe it to me, so I have to buy it online. I hear about about all sorts of other people getting it prescribed for the same problems I have even though I stumbled on this on my own. It's very frustrating to have less informed doctors, but I guess that's what I get for being on medicaid.

It's also very popular MAT for methamphetamine addiction (for the doctor's treating not so much the patients). When I went to treatment at the VA a few years ago for my heroin/PTSD I saw a lot of dudes that were there for methamphetamine and everyone was either currently on or had taken this medication.

Interesting I'm prescribed this so it's very informative. Can you make a video on making the nicotrol inhaler liquid and the nicotrol nasel spray. In the US it's persciption only since forever mean while Cigarrettes with 100's of chemicals and vapes are fine but cause sever health repercussions. I think it would bring awareness to an alternative that needs to be less regulated by the FDA so people can be healthier and not pumping money into pharmaceutical companies pockets. Such a straightforward synthesis! It's a shame how much the drug used to cost in America before going generic (and litigation debacle with Cephalon IIRC). I'd be really curious to see an armodafinil synthesis / resolution. Looks like that involves synthesizing the methyl modafinate, hydrolysis to modafinic acid, and enantioselective crystallization.
FYI you can get Flouro-modafinil legally to "research" in the US

Does this create a racemic mixture? If so, it would be cool to see a dynamic kinetic resolution. Afaik the R enantiomer is the biologically active one

Ohhhh, Phenyl Acetic Acid. That one is really hard for people to get here in the States. I mean, I have several pounds of it saved back, but that took me a long time to find a good supplier who wasn't Sigma. I absolutely love the smell of it though! It's not quite like honey, but it's so close that if you didn't know any better you would swear it was. Lol!

You mentioned you might methylate the amine in the future. Do you know anything about the effects of methylating the amine? Will it be maybe faster/stronger acting because of better blood-brain-barrier crossing or might the potency go up or down?
Probably acetylating it with AA would help pass the BBB easier as a potentiator.
Just so everyone knows this a CIV drug in the US (same schedule as benzos and barbs). Don't try to make this if you live here.

@Tyke Actually it was once banned. It was explicitly removed from the german illegal narcotics list in 2008
I only knew of it being controlled in the US tbh, in my country this is over the counter (and funnily enough, even though this is supposedly also a nootropic and also can be used for pulling all-nighters, a survey found that it's Methylphenidate, which is actually controlled here, that is the go-to for university students)

Great video! Can you start with half the amounts mentioned? I only had 40g of chloroacetamide so started with 32.5g thiourea, 65g benzhydrol and 165g distilled water. Added about 65g hydrobromic acid. I think the solution turned quite clear but not bluish. Followed the rest of steps halving the amounts mentioned. After adding the chloroacetamide I strirred for 4h, in the end it was a yellow slurry, like a lava lamp, yellow slyrry rising and falling. It was mixed and filterder at 70 celsius like the paper says. At that temperature (70 deg.) most product went though the filter easily and only 4.5g white soft product remained in filter. Filtered the yellow stuff again at 60 deg. and got more product (49g yellow stuff). Prodeeded with rest of the steps with the yellow product. After adding the H2O2and recrystallation I was left with 49g of yellow a bit crystalline looking product. After boiling in methanol and little bit water there remained part that didn't dissolve no matter how much methanol was added. Put it in fridge but it had too much methanol to recrystallice so boiled most off and got end product 33g of a bit crystalline yellow stuff that is hard to get off the labware. It has no effect on the human. Any ideas as to what went wrong?
Of course you can start with half the amount. it took me three times to master this synthesis so i´d say just try again and be even more careful.

This particular compound has a biologicaly active crystaline structure that needs to be obtained by a specific recrystallization procedure. I did the same erowid method and then I found the recrys method in some patent, the bioactivity is incomparably better.

Modafinil is such a cool drug. You can eat and sleep while taking it, gives zero euphoria or jittery feelings.

@@ThoughtMachine1 Even when I take 400mg, it's not overly stimulating. It's like caffeine - only it doesn't make you jittery. I don't know why it's controlled substance, but I suspect it's abusable. I know someone that said they take huge dosages. I wish people wouldn't do stuff like that. It makes all the authority wanking control freaks feel justified in controlling it.
I tried a version of modafinil that was sublingual. With that I did feel a slight euphoria right at first.

Fmri shows enhanced nueral activity but not of Mu receptors it may be the enhanced activity that made it feel euphoric but you're safe, it won't activate anything euphoric of the type that leads to abuse and addiction

Interesting and fascinating chemical. Quite cheap the generic India made one. Be careful if used in combination with other drugs. Combo with low dose amisulpride gave me a tremendous headache and weird dual tone tinnitus.

What do you mean don't add too much HBr or we destroy the thiouronium salt? Is it something specific to benzhydrol or TU salts in general? Asking since I tried to make other thiols like this and in one attempt I got fucking sulfur lol. Though I did use KBr and H2SO4 instead.

I took Modafinil a few times.
As a stimulant, it wakes you up, but does not cause you to be jittery like caffeine does.

I found this drug by mistake and asked my doctor if I could try it. Would love some help to understand why it fixes me. I can't eat foods like red meat and to much gluten.
Citric acid from oranges left in water also helps me.

I looked up your condition, that's basically how I feel like my body is lacking oxygen. I get tired really easy and down because of it. I'm better in summer?? The modfinal helps keep me above the struggle line, it just makes you feel functional.

It began in Hugh school that I couldn't even do my homework after school because I was too tired. My parents think that it is just in my head, but even themselves sleep a lot. I sleep more I'm 19yo. They are 47yo. I think that I shouldn't feel like and 47yo.
So I'm currently in nursing school and in 4 years in medical school. If a doc. won't help me I plan to do it myself, even if I have to produce it by myself in a laboratory. I want to be away.

i didnt even realize it was legal in germany lol, i remember trying to make adrafinil because i didnt have any proper way to make the chloroacetamide without crying lol.
also did you do this all in one day? i remember being limited by time and all my products dimerizing and tarring up into disulfides if i let them sit for a week.
the entire prep if we exclude the time needed for drying the reagents and just waiting to continue because i did not have time sums up to about two days. if we include the time where products dried in the vacuum chamber it is four days. may eventually make adrafinil and modafinilic acid. it would be cool to make modafinil versions with other substituted amines and this is easyiest from modafinilic acid chloride plus primary or secondary amines.
@@THYZOID if you do make adrafinil, please share your process, because i had a damn hard time purifying the intermediate after ester amidation with hydroxylamine. (CRL-40,941 would be a cooler pick, especially since youre sponsored :P)

i would be weary of taking fluorinated drugs daily. while conventional reddit wisdom is that C-F bond is unbreakable, this is not true inside a biological system. in fact, patients taking certain medications long term --- voriconazole and ciprofloxacin for example --- must be monitored for fluoride toxicity. notice, this is even despite the fluorine from voriconazole needing to be liberated from an aromatic ring(!) (20mg of fluorine per 400mg of drug were liberated). as flmodafinil doses are in the 100s of mgs, it is probably not a good idea to take long-term.

Science would not be where it is today if not for many years of intensive research, testing, reexamination, theory etc. From what I understand , for many keystone scientists and researchers across all disciplines, it was not uncommon to use stimulants such as amphetamine in order to meet the demands of a thesis or course of study.
It would be faster for me to build a lab and make this myself than wait to get admitted to the sleep clinic. I hate healthcare in my country.

The dose makes the poison. Just because something is made from chemicals does not mean it is poison. Everything in nature is made from chemical compounds. I just cannot incentivise people to try their self synthesised modafinil.
I have a request as the information I have researched has proven unreliable and unsafe. How to successfully remove codeine from prescription medication that are mixed with paracetamol, many people say doing a cold water extraction works, however the research I have done from hospital admissions showed paracetamol was still present and therefore harming the kidneys and liver. Is there a safe method to extract the codeine from the paracetamol.
