I will really suggest a child theme setup for that instance, a valid concern indeed that's why we have always encouraged child theme setups, parent theme updates doesn't effects child theme's code so all the customizations made to the child theme remains safe, we will have a video about how to create a child theme very soon in this channel. I tried to modify the page which display the event. So I copied the file "content-single-event_listing.php" into my Divi Theme (wp-content/themes/Divi/wp-event-manager/content-single-event_listing.php). But, modifications of this php file doesn't appear on the page... Did I make a mistake ?

Unfortunately, that is part of the functionality of the plugin. You can remove it by overriding the template or by personal customization.
How to protect the override code , if the theme is updated by client(administrator), if I add new js files or css files?
