I have made the cheapest laminar flow hood possible. Simply using a cheap air filter purchased online, I was able to achieve clean plates in a very dirty environment.
This design is simple, and worked really well. With a bit more effort you could have this device turning out clean plate after clean plate.
This device works by blowing filtered air over your workspace, which prevents contamination from landing on your petri dishes. In turn this creates a clean area to work in front of. I could have improved air area this device was working in, but for the purposes of this test I didn't put much effort into cleaning the area or room prior.

You should take the filter out of it and run some smoke/fog through the intake and see what kind of vortex areas are not cycling clean air in the box section.
i would hope fog instead of smoke haha,
but really, this is an example of overkill, you wont get any contamination from a corner. contaminate would have to like, you know, get to the corner in the first place, and then survive there indefinitely,
meanwhile I have 6 fruiting unmodified tubs without any still air box or air purifier or flowhood or any of that fancy crap,
because we've been farming mushrooms since well before technology,
I deal with contamination sometimes, but so does everybody :)
I say this because i knew a guy who would get aggressively angry about sterilization, he was so whiney about everything in his grow room. might as well have installed a full body sanitization hallway and hired security.

More labor to build, much higher cost, and more time wasted to end up with a lower success rate (25%) than what can be achieved in a SAB (upwards of 90%)
Great video visualizing why the experienced, well informed, and most successful cultivators don't advocate these contam fan contraptions. SAB, flow hood, or FFU.
*All three still require good aseptic technique for the best success rates* but the baseline is already higher for anyone new and trying to learn. Save yourself the frustration, folks.

I think that part of the reason that you had contaminates is because the air isn't actually laminar - its just clean air blowing through a box with no pressure. I think what would probably make the work close to 100% is to have a prebox. So the purifier blows into the prebox (which could be much smaller of a box than the post box) and have a cheap regular air filter just so that the air pressure could build up some and take away the chance of contaminates in the corners, then as the air comes from the prebox, through the air filter into the post box, it would actually be laminar and the inside of the post box would have much less of a chance at having a contaminate in it. That along with the other things that you mentioned like vacuuming the carpet, allow the air purifier to run for an hour before you start, not having the big fan off camera running, etc. I'd love to hear your thoughts on it.
Or perhaps just put a regular cheap HVAC filter on the outside of that box, that way you'd get pressure built up inside of the box and it would come out as laminar.

A cheap filter wouldn't produce much back pressure, so you wouldn't get laminar flow. Even cheap HEPA filters can't do it, you need the big thick ones.
The reason this isn't working is a combination of not being laminar, and the fact that air purifiers don't put out sterile air. They leak air around the filter, and they often have a bypass behind the filter that sucks in a small amount of dirty air, so the sensor can detect if the air is dirty or not to turn the purifier on or off.
@@LarsLarsen77 silicone around the edge of the filter then simply cut the silicone with razor blade to switch filters? Random construction worker spewing shit. The air leaking around the filter is the only thing that's stopped me from doing this same thing with a dry cleaning bag with two slits cut for arms half way down the tube and a small exit hole at the end. Is it laminar? Nope. Could the positive pressure decrease my contamination rate having 5dogs, 3 cats, and a squirrel?? Idk, you tell me. Lol
@@LarsLarsen77 and no it wasn't my original idea. The air getting around the filter was just a flaw I noticed with these cheap purifiers.

If you can do it in a room with dirty carpet and a fan running, I bet I could make this work even better in my freshly sanitized bathroom. I started with a SAB before I knew about laminar flow hoods. I had good results but thought adding a hepa unit like yours might be even better. I eventually tried 3 different hepa filter units in various configurations (we live in a 150 year old cabin and have allergies). I learned that you must reduce turbulence by ensuring that the air flow is channeled over smooth even surfaces, no bumps, wrinkles, niches, etc. My current hepa setup is simply a rectangular frame (cardboard) fitted inside a large plastic bag to create a wind tunnel. The tunnel matches the dimensions of the hepa unit: 22 inches tall and 10 inches square, just over 2 feet in length. The outflow is from the side so the unit is laid down on a table and the bag is well sealed around the outflow. I mist the interior with alcohol, let it rest a minute or two, then start the unit about 10 minutes before I begin working, I also dampen a towel and slip it inside the tunnel, covering the entire bottom (no wrinkles!). I've used this for a couple of months and my rate of contamination is very low, certainly 99% clean. I did an initial test with open agar plates (5 minutes) and only the plate on the left showed contamination. Since then I've done agar-2-agar, g2g, LC to agar, clones to agar, and a bunch of other stuff. In broad terms, I've worked at least a hundred petris and several dozen grain jars in front of this thing. The tight working space is awkward -- 22 inches by 10 inches -- but on a rather low fan setting this unit puts out serious wind. In fact, it makes my hands very chilly. I sit to the left side, which allows most of the air to leave the tunnel without creating additional turbulence and possible back drafts. This is clearly not laminar flow but for the sceptic all I can say is... it works!

I clam shell my covers open, drop the inoculum and close the lid. The dish is open for only one or two seconds. Five minutes made for a great demonstration.

I'd imagine this setup would work great as a flow hood/still box hybrid. The positive pressure inside would ensure its only filled with clean air. Thanks for sharing this experiment!
It doesn't work when you have such wide dimensions around the unit. He's just creating vortexes. Better off using a bag that will conform to the volume of air and create real positive pressure.
I mean when you break it down. You had one or 2 contam points after leaving the plates open for 5 minutes. I’ve never kept a plate open for more than 30 seconds. I find that damn near impressive. Better prep, and realistic settings would make this a much better experiment. I mean, you can make the argument they got contaminated, but limiting exposure and pre prep are both extremely vital to clean samples. Awesome video, loved it

Interesting! If you decide to do this experiment again (along with all the things you mentioned in the video), it would be interesting to see how far away (or closer) from from the filter you could get and still get a clean result. Maybe have plates spaced every 5 inches away, starting from the filter output till a few feet away? Also, it would be interesting to see how this compares to a still air box, since those even the cheaper alternative to this. Thanks for the entertaining vids and hope your arm is healing up OK!
That would be! Next time I would use it in a room like my clean room, I think I would get 100% clean plates If i used it in there.

A friend of mine converted a similar sized SAB into a downdraft flow hood by adding one of those air cleaners to the top blowing down. Works a treat with the air exiting the rectangular cutout at the bottom front of the box.
I was envisioning that same thing after watching a video on how to build a Laminar flow hood. Maybe add a UV germ killing light too.
I was thinking about doing this, thanks for the test run. it sure beats $1,000.00 USD + Shipping for a 2'X4' laminar flow unit until I can make a bit more cash off my mushrooms.

I'd love if you would revisit this idea! Perhaps make the "hood" smaller? I think turbulent air in the box would be the main drawback of your first design (maybe check the flow with a stick of incense). I would opt for a box that is almost exactly the same size as the air purifyers outlet to make sure the air stays laminar all the time. Also crank it up to setting 3 and see what happens.. It might keep the air laminar for longer.
Sure a smaller hood is not ideal... But for most hobbyists its not a big deal if you are only able to work with one plate at a time... As long as its sterile!

I went to an urban mushroom farm here in Greece, and the people there were very generous with their advice. They showed me their whole setup and explained it. Now, one thing sprang to mind when I saw your video was a potential improvement that I saw in their lab.
Instead of keeping the box open and fiddling around with petri dishes in open-air environments, they instead had made a closable glove box with the "arms" of the gloves made of Tyvek (semi-permeable material). It was a tote exactly like the one you have here, but with the lid closed and sealed with a squishy rubber string thing around the rim. They pumped air continuously while working into this tote through a small plastic pipe, which escaped (along with particles) through the Tyvek.
If you retrofit the cap on this box with permanent gloves and make a slot with Tyvek or something similar on one side, then load it up with your dishes, run the air pump for some minutes to purify the air, and then stick your hands in, perhaps you'll have more success. Remember also to put your isopropyl alcohol in there before closing the lid.

That would be a good idea, but this was just a proof of concept. I use a larger flow hood in my clean room. If you left this unit running for 4 hours in that room prior use, it would perform a lot better.

You don't need no glove box. It has been proven time and time again by whole lot of growers and cultivators that attached gloves only make things worse, because attached gloves are fiddly, and the less you fiddle the less contams you spread around. LFH's work, SAB's work, no need to invent the wheel and complicate thing more than they are

Great video! I wonder if some resistance might help. Maybe there’s a stock size air filter that will force fit in the tub and provide more even output. Then you will want to turn it up to 3.

I think a cheap improvement would be to block the top half or so with a removable panel. The air flow through the remaining space would be greater and eddies would be reduced. The panel can be moved aside when large items must be moved in or out.

I'm thinkin using 2 tubs inside one another and sealed. The same setup in the rear with the blower, and holes in the bottom of the forward tub would cover a larger area inside the tub and probably blow harder.
Maybe even a home ac filter between the two?
No expert, but I reckon the plates in the box have slightly more contam because of the wider opening, as you said. The displaced air in the corners has to pull in air from the front, which causes more turbulance inside the box than in front. Pretty much what you predicted. Really interesting to see this though. I'd say a small, flat filter with a blower is the way to go though in terms of value for money.

@@alr2989 so far zero contam but thats mostly with syringes still and g2g which is pretty risk free to begin with. Ive been pretty lazy about prepping some agar but ive got today and tomorrow to work on it, ill try and have a better update by the end of the week. Also im using a different $40 filter and a larger tub so just be aware my results might differ from his.

@@alr2989 smart move, i tried to jump off into the deep end too much and wound up with a big tub that only partially fruited. definitely think pf or bag rice or any of the smaller batch teks are the smarter starting point.

Actually had a similar idea but inatead just use SAB, cut hole on one side for HEPA filters, other side gets a hole for a intake or extract fan, put that fan in there, seal all caps the box has so air can only go thru the filters and there u go, fully filtered and actual flow instead of air circulating

How do you innoculate bulk substrate in a SAB?
I have had incredible success with a regular box fan and using furnace filters gorilla taped to it. I have a pre filter on the back of the fan and then I have 2 stacked one in front of the other on the front with the best quality furnace filters you can buy at wal-mart or some place like that. The air flow is really close to laminar if not legit laminar, it's laminar enough to work really well for me, I know that much.

What material do you use for a prefilter, is it just some cloth on a frame, or a manufactured item?
@@eclogite I use a cheaper furnace filter for the pre filter. One that allows a little more air volume to go through it.

Wont the air just blow out the front rather than filling the box properly including at the edges? Perhaps adding some washable G2 nylon filter material in front of the airflow could help build up the static pressure a little (hence filling the box) to create better laminar flow. As long as the purifier has a strong enough airflow.

This is a good video. Setting 3 on the fan would give you more airflow right? Closer to 100+ cfm?
Try using 2 air purifiers one as your flow hood and one in the centre of the room away from the flow hood while the main one in the centre of the room is inhaling most of the contam in the room

Certainly isn't. These are called "contam fans" in the experienced cultivation circles. SAB=cheaper, easier, much better success rate. Have you figured that out in the 7 months since this comment?🙈
@@brantlyrobbins2718 well the first process is learning to be sterile. This would help to a degree with someone starting. I've been doing this for way more than 7 months and can do open air w/o contam, so yes I figured it long before this video. SAB is still the best to learn sterile technique in my opinion, also it never hurts to experiment. None of this would be possible without experimenting.

I use one similar to this to positive pressure my lab as well! I mounted it though a wall lol...
I believe the contamination is because it wasnt laminar flow , thats why the plates that were outside the box were cleaner , try leaving it on for 5-15 minutes and reduce the air speed .

If you had another heppa filter or filter of some sort after the air purifier but inside the big box before the opening it should form a positive pressure if my brain is correct. Im going to do this build myself and find out! Cant wait!

I use a still air box in a room with plants and carpet and I have a very low rate of contamination.
I found a similar video, but they used a dry cleaning garment bag taped to the output to make a MUCH bigger work space. Just cut a couple holes in the bag for your arms, spray everything inside with iso, and you're good to go. It may not be laminar, but the air inside will be super pure as long as you're using a true HEPA filter.
Price tag for these things are a bit pricier depending on where you live. Had to drop just under $120 USD for the unit plus 2 HEPA filters (it came with only an allergen filter stock)... but compared to a flow hood ($500ish for that one Chinese unit on amazon, or 1000+ to source your own filter and a fan strong enough to generate laminar flow, plus build the housing), it's hard to complain.
I did a control plate, exposed it to the air inside the bubble for about 10 seconds, sealed it back up. 2 weeks later, nothing visible on the surface of the agar.
You dropped $120 for the blower only or both blower and 2 Hepa filters?

@@WatchTLCCRMOFFICIAL Target, but the filters are cheaper on Amazon. Some states, air purifiers are a bit more expensive due to stronger standards and regulations, too.
If the garment bags were off a roll or otherwise new and unused, might they be sterile? Enough? I hate the ISO makes me cough and gag and I still haven’t figured out how to flame sterilize without blowing myself up.
You can actually use a poly bag. tape it to the the purifier, let the bag inflate and then take your scalpel to cut arm holes and one access hole. 😉

I want to see this compared to a still air box. At over 70 plates I've only had one contaminated plate in my still air box when I'm working in my my living room. I typically run a cheap ionizer inside the still air box for an hour before I get down to business , something I believe is helping to pull contaminations out from the air and sticking them to the surfaces of the box so I can wipe it off with 70% iso.
I wonder if this diy flow hood creates spots of turbulence that pulls in contaminated air along the sides of the plastic box, something that wouldn't happen in a still air box because of barely any air movement.
The problem with a still air box is you cannot innoculate lots of bags inside it. With this you can.
Iv made one similar but i used 2 filters one flat one round and a small ducting fan iv never had contamination yet cost $100 👌

So I was thinking of this same thing. But I think creating a positive pressure environ inside a box means the amount of air flow does not matter. I would close the end and put a couple of holes in it like a clean box. Let the purifier run for a few minutes with your stuff inside. That's the angle I was going to take, mate.
I seriously ghettoed mine. Bought 2 of the purifiers he's used here, larger tub, sellotaped the life out of the purifiers to hold them in and sellotaped the front up leaving just a rectangular space in the middle for my hands. No idea how it has worked as yet as not long inoculated, done agar making and transferring , will find out soon. I had been using a SAB, but honestly im just a very clumsy erratic person and for me it's hard to be as delicate as is needed. My thinking is, it's very well taped at front, seems to be very consistent flow out, put multiple tape strips on the front space to check if they flap around and they stayed consisteny blowing outward. Hopefull mean i have enough sterile air being pushed out the box to at least make it better for me than the SAB was. I can live with some contamination i just need better results.

OMG Your the only one besides me who gets it. You don't need a tornado, just a top quality filter and positive pressure!
Use a dry cleaning bag or a large trash bag. Duct tape it to the air purifier. Cut armholes in, probably further away from the air purifier. Turn the air purifier on and it will create positive pressure. You can then spray the inside of the bag with iso.
Or just use a SAB and save yourself some headaches. SAB work is perfectly sterile enough if you're doing it right.
I'm going to do this but I'm going to cut arm holes in it like a still air box and put the fan in the top of the tote
will this same set up work installed upside down like hanging from a low ceiling????
How did this work out? I have a couple Holmes air purifier filters coming, I might try this out.

Could that contamination be from you talking in the direction of the plates?
Did you do a follow up with more thorough prep? It didn’t really sell the idea more than using a DIY still air box.
