Do prospective mitochondrial proteins (for example) contain an ER signal sequence as well. So the protein can be transported via endomembrane system to reach the mitochondria. Or, does the protein just have a mitochondrial signal sequence which is transported from the cytosol to the protein translocator of the mitochondria without going through the endomembrane system with the start at the ER? Or, do both of these occur for prospect mitochondrial proteins?

For the protein to be targeted to the outer mitochondrial membrane it is basically the same mechanism, only there is another transporter called SAM that is also located in the outer membrane that will transport the protein into the membrane. The protein will go from TOM to SAM rather than TOM to TIM23.
Protein found in intermembrane space contains an N terminal matrix targeting sequence followed by cleavable hydrophobic sequences that blocks complete translocation .How sir?
