  • Deoxyribonucleic acid is a molecule composed of two polynucleotide chains that coil around each other to form a double helix carrying genetic instructions for the development, functioning, growth and reproduction of all known organisms and many viruses. DNA and ribonucleic acid are nucleic acids.

Sir, there are 10 %adenine among the bases in a DNA fragment measuring 10.2 nanometres length. The no. Of H-bonds in the DNA fragment.
    Answer is
    3.4 nm - 10 bp
    10.2 nm -?
    =30 base pair (bp)
    10% of A means
    20% of A+T in 30 bp
    So, 20/100*30=6
So, 6 base pairs of A+T
So, no of H-bonds of A+T is
    6*2=12
    So, 12 H-bonds of A+T
    80%Of G+C bp's in 30bp's
    So, 80/100*30=24bp's of G+C
    So, 24*3=72 H-bonds of G+C.
    So, 72+12=84 H-bonds.
Now, if the answer for that question becomes a question like this.
A DNA contains 84 H-bonds. It contains 12 Guanine bases. Find the no. Of Adenine, Cytosine bases.
The answer will be like this.
    =12+12 base pairs of G+C
    So, 24*3=72 H-bonds of G+C
    84-72=12 H-bonds Of A+T
    So, 12/2=6 bp's of A+T
    6/2=3
3 means 3 Adenine and 3 Thymine bases.

In the same way if I solve the below question.
If a fragment of ds-DNA has 135 H-bonds. It contains 23G-bases. Find the no. Of Adenine and Thymine bases.
      Answer is
      =23+23
      =46 bp's of G+C
      No. Of H-bonds of G+C
      =46*3
      =138 H-bonds of G+C.

      23 pairs of G-C x triple bond = 69 bonds
      135-69=66 bonds between A-T/2 bonds per pair = 33 base pairs A-T
The answer: this dsDNA contains 33 base pairs A-T

Thank you for your videos. Base pair length is my biggest problem now. I still don't understand how to determine base pair and size. The samples of the kit i use give the base pair of the gel around 118bp however the marker/ladder i use range from 50-10,000 bp. Now on my result, the DNA bands are little below the lowest marker band. How can i determine the base pair length and size of such a result. Thank you in advance!

    • @GeneticsLessons
      @GeneticsLessons  7 років тому +2

Oh, that is simple - you have to run a polyacrylamide gel (not agarose) and reference-ladder between 100-150 bases, then you could find the size of DNA fragment exactly. Good luck!

If the n=8 in plant cell, what is the no. of tetrads, no. of sister chromatids, no. of centromeres in pachytene of meiosis...?

    • @GeneticsLessons
      @GeneticsLessons  4 роки тому

if n=8 than 2n=16 and during prophase I chromosome number will double 4n=32
number of tetrads 32:4=8 number of sister chromatids 8+8 and 8+8 or 16 of one kind and 16 of the different kind (this question is tricky you can answer 16 of one kind or 16 of one and 16 of the other) number of centromeres 16 - one per a pair of sister chromatids.

Если можно у вас спросить, как посчитать наиболее вероятное число пар оснований в последовательности узнавания, если при расщеплении ферментом образуются фрагменты средней длинны 1,048,500 bp.
When DNA is digested by this particular enzyme, fragments are average 1,048,500 bp in length are produced, what is the most likely number of base pairs in the recognition sequence of this enzyme.

If we divide 10 base pairs conatian 9 equal division so 3.4 divide by 9. 0.37 so 0.37 is the distance between consecutive base pairs?
