Idk if this will help anyone else, but i only just realised. If you tap on the boss, it will tell the AI to focus that rather than the lil scorpions by it's side. May be obvious for some but i legit didnt know that and it shot my score up (obviously xD )
I thought that you needed normal shaltear in the team and alive to get the defense down to work? Isn't the first part of the description the passive for the character combo? I've been confused on this but haven't tested it myself.

Moman, Maid Albedo, Valkyrie Shaltear and Trail Pandoras Actor got me 23k without any buff scrolls.

I just watched the clip again, and you never trigger defense down with that Supreme one ability, only high-tier heartbreak.
The "defense down" debuff is a bonus effect when you have Shaltear (warrior) on your team
