Fun little thing I learned about a while ago is that majority of comedy is about subverting expectation. It's why the rule of 3 works; the first instance isn't anything, the second establishes a pattern, and the third is used to subvert that pattern. It's why anything that starts with "I'll tell you a joke" isn't funny, because you're expecting a punchline, and why non-jokes can sometimes be hilarious.
You left out a major factor, yes pun intended since you are after all, a mathematician. You forgot to factor in the fact that laughter is contagious. I laughed at your "3 prime numbers" joke not because I'm a mathematician but because at 3:43 the expression you gave is of legitimate humor indulgence. Basically, if someone cracked even a bad joke at me, and legitimately laughed at it themselves, no matter how dull of a joke it is, I wouldn't be able to help but laugh along like a cackling hyena..
for some reason i watched this video thinking about the actually mathmatics behind finding out perfect comedy routine. And then relized that i'm quite certain alot of comedians use this. I think im going to research and find out if certain comedians actually time things specificity, and see if there's a average timing for certain jokes. Shouldn't the axis labels at 2:23 be switched? "Punchline Delivery" should be on the X-Axis since it's the independent variable while the "Audience laughter" should be the Y-axis since its the dependent variable.
Comedy can not be scientifically analysed, It is more about the character of the person telling the joke, their accent appearence and body language also play a huge part, I agree timing is important, but some people are just naturally funny.

I just laugh at my own jokes. I stimulates laughs from others, whether at the joke or me. but it works! :D
