    I just started pramipexole a few days ago after 30+ failed medications for refractory MDD and ADHD. I've tried everything from 12 sessions of ECT to macro and micro dosing of psilocybin from homegrown mushrooms, all with no help whatsoever. Fingers crossed.

      @@maikedasilvasantos7461 It's working great!! I'm now taking 3mg each day: 1.5mg in the morning and 1.5mg at night. It has CURED my depression. Seriously. It is saving my life.

      How long did it take you to notice a difference on nardil? What dose were you on? Does it give you back the energy and motivation to get back to work/school/hobbies/social life?

    I have had major treatment resistant depression for twenty five years.tried all the antidepressants with little help.I saw this video and asked my doctor for pramipexole.the doctor said no.any one know a legit online pharmacy where i can get it.doctors should be willing to try things till something works.

      +bob malone i've ordered from buy-pharma. co, they have pramipexole, i believe its made in india. The meds ive gotten from them have been legit.

      @@capitalist4life i appreciate that but unfortunately i'm in the uk where we have a formulary based prescribing system and so it's very hard to get even a psychiatrist to prescribe something not on the formulary

      Nitroblast have you tried a TMS? Look into Fisher Wallace stimulator and of course the pramipexole, which will help give u back some ambition and drive. (Treats anhedonia)

    Low dosage of flupentixol helps with dopamine related depression for me. I guess it is not mentioned here because US does not approve this drug.

    I have been on pramipexole (as discussed in this video at 45:32) for about 6 weeks now. I have severe TRD and I would say I am about 25-33% better since starting pramipexole. I have cut back to 2mg (from 4mg) . This is the first med out of several in the last three years that has done anything at all for me.

      I am now on 4.5mg of Extended Release and that has made a big difference in lowering the side effects of pramipexole. I would say that I am about 25-33% better since starting prami. I haven't felt any real improvements in many weeks now, however, I also have not had any downturns or relapses. It seems that I have entered a maintenance phase and I am not sure that I will see any more positive surges. I have been on the drug about 8 months now. it was at the 3-4mg doses before I noticed any positive changes, smaller doses had little to no effect for me.

    I wish studies like this were done for people like me who are 18-30. I’m about to be 26, been on meds since I was 14 nothing has worked and my doctors have given up.

    For the old guy with treatment resistant it is possible he may be getting early symptoms of Parkinson’s since early symptoms are usually depression and since it is primarily dopamine related symptoms

      What???? he died? Ohh no! i just came across this video and watched the entire presentation discussion here with him on here. If he has passed away, that is so sad!! I would have loved to have contacted him, as I am laterally desperate, myself.
      9 Years in particular (but suffering for longer, just more as years go by) trialling all different antidepressants and mood-stabilizer medications, psychological therapy, TMS, and other lifestyle change attempts. Suffering from Treatment-Resistant Depression or something else with Depression.. I have Anhedonia now for the last couple of years, i have suicidal ideations all the time (but never attempts... yet) , Irritability, sleep issues, skin-picking/self-harm, some alcohol misuse previously, but trying very hard to abstain or really limit consumption...
      39 yrs male in Australia - also with birth disability.
      Current Psych does not want to try MAOI's or Ketamine or ECT, or VNS - yet. I dont either, only because of the terrible effects or side-effects. I have never heard of Pramipexole, until now, watching this......
      Is it available in Australia???
      I am desperate to try anything that will give me some feeling back, my life back, some level of comfort and stability to feel some enjoyment again! I would rather be dead at this point!! But i know that, that is not the answer, but i keep getting worse, and do not know why anymore, when i have tried things on my own end to to help myself.. And still feel terrible and lifeless... And if the Psychiatrist's here do not do anything different than the standard treatments, and , do not know about Pramipexole , then , what do i do???
      Sorry to hear he has passed away! He really does sound like he was a high intelligent and caring guy, and knew of different treatments and willing to try it on his patients..... Sounds like he was a lovely guy!

    The patient you spoke about 26 min into the video. You mentioned he had prostate cancer. Was he being treated with finasteride prior to getting surgery? Robin Williams was on that medication for hair loss treatment. I fear it contributed to his suicide. I can tell you first hand it causes VERY serious depression. That medication breaks something in the brain and it stays that way well after stopping the medication. There is research underway in Italy that shows the drug greatly impacting neurosteroids levels well after stopping as well. Some of the same neurosteroids deficiencies recently found in MS. The stuff is pure evil in a pill. I would rather die from cancer then take that medication. I really wish more doctors were aware of the power that medication has to induce these problems.

      Yes. Finasteride anxiety is quick and pretty bad. It’s a real shame that they have not developed an analogue without BBB penetration. There is very little benefit to having it active in the CNS

      Good luck. That was a formula used by natural herbal co. Big pharma bought it...that formula got my dad off zoloft and a sibling off prozac.

    Each Maoi is unique even though they are same class. One is excitatory (parnate) vs inhibitory (Nardil).
    This is a brilliant dr that thinks outside the square!!
    I just wish more so called psychiatrist n dr were like him.

      Try contacting Dr. Fawcett and see if he'll write a letter to your psychiatrist describing his strategies for treating anhedonia. Dr. Ken Gillman is another eminent psychiatrist who understands the importance of dopamine, he responds to emails and has an open letter on his site you can bring to your doctor ( psychotropical.com/open-letter-using-monoamine-oxidase-inhibitor-maoi-drugs/ ). You could also print out the studies mentioned in the video on pramipexole and MAOI treatment and bring them to your next appointment.

      Same. I've told more than one Doctor several times that I wanted to abstain from masturbation and porn addiction, which I have done through sheer will power, but after conditioning my brain with that GARBAGE has left me with anhedonia and insomnia, erectile dysfunction, short term memory loss and poor focus, and subsequently acute depression, suicidal ideation. Most Doctors do not give a shit about their patients. That's what I'm begining to think.

      ask about trying wellbutrin, it helps low dopamine, and doctors generally aren't too hesitant to try it. sometimes they might wanna try an ssri first but if that doesn't work, wellbutrin is a pretty common second attempt

    I've treatment-resistant depression and was looking for solution. After finding this one, I immediately discussed pramipexol with doctor this morning (March31,2021). His response was not positive as he was of the view that few successful cases couldn't be the benchmark. So he doesn't advise it. I desperately wanted to try it as I have been testing SSRI, SNRI, and TCA, for 20 years. Nothing helped. Still down and restless with mental fatigue and low energy, low self esteem, sleep problems, anhedonia and many more issues. What should I do? Should I discuss this with another doc or this medicine is no longer relevant as this video is 7 years old. Response from Brain and Behaviour research foundation would be appreciated. Thanks.

      @@tayyebafridi I ordered it from overseas. The side effects can be pretty bad but go away with time. The one side effect to watch out for is compulsive behavior. That can be VERY dangerous.

      Yeah it’s weak and has a high side effect profile. I get horrible tremors on 450mg/day and my vision blurs really bad.

      It mostly raises noradrenaline which is probably what the side effects are from. Noradrenaline is similar to adrenaline.
      Insomnia, sweating, overstimulation etc

    With such a high dosage target and only once daily dosing, wouldn't it be wiser to use an Extended Release formulation of pramipexole to avoid substantial daily blood level fluctuations ?
    Dr Fawcett never mentions the subject, so I guess he's using the Immediate Release formulation.

      @Katie R Yes I have, for approximately one month, but I couldn't ramp up the dosage very high (1mg is the highest I could get). After a while it gave me intolerable insomnia that didn't abate even with a dose reduction, so I had to stop. I didn't feel much of a positive effect, just some sort of unpleasant stimulation and a buying compulsion that I had to repress.

      @@gregorymalchuk272 As for me I would say no, but again, I didn't tolerate it, so I couldn't take it for long enough on a dosage high enough. By the way, I read somewhere that intolerance was rather high (around 20%). That's a fact that Dr Fawcett doesn't emphasize so much about his wonder drug...

      tardive dyskinesia is caused by anti-dopaminergic drugs (risperidone, haloperidol, etc.) not dopaminergic drugs like pramipexole

    D2 receptors have got to be involved in depression. Abilify, a D2 agonist, works incredibly well for me.

      capitalist4life it’s partial d2 agonist meaning it blocks excessive d2 stimulation but slightly stimulates it , abilify (aripriprozole) is also of serotonin receptors 2 antagonist which has antidepressant quality and a serotonin 1 receptor agonist which also has antidepressant quality
