Making a Hypnotic Sedative from Acetone

  • In this video I make an old hypnotic anesthetic agent called chlorobutanol from acetone, bleach, and potassium hydroxide.
    Note that this chemical is NOT a recreational drug, and I have yet to find any examples of someone ever trying to use it as such. This was used nearly exclusively as a medical anesthetic until its use was discontinued due to its major side-effects which I discuss later in the video.
    As always, do not attempt anything you see except under professional guidance.

      It doesn't work like that with super long acting benzos or phenobarbital. The high is still like 20 hours, and yes with phenobarbital it's half life is like 20 days, you are not feeling the phenobarbital for 20 days more like 10 to 12 hours yet you will test postive for weeks after it wears off.

    Some time ago i worked in a factory that (next to other stuff) made this bloody stuff in the tons range
    Gave us all sorts of problems with the centrifuge and discoloration and quality control
    Edit: The reason you can't find any literature about the reaction mecanism is, that the mecanism is not fully understood yet. I searched for it for the better part of a year and could only obtain some information on side products and reaction conditions
    Never again but very nice video once again

    The half-life is long because it's a tertiary alcohol, which cannot be readily oxidized like chloral, or trichloroethanol, or ethanol.

      Yeah thats what I figured. I honestly am not sure if your body can even oxidize it at all, I think I read somewhere that it is metabolized by glucuronidation or sulphation, and then eliminated renally.

      @@integral_chemistry I wonder if it could worsen liver or kidney function if there is already a reduced function of those organs. With its half life, I wonder if it could cause a stead and long term decrease in renal function for someone that has CKD and does not know it.

      @@user-nu8in3ey8cif it’s that hydrophobic, probably not. Kidneys typically filter out H2O soluble compounds, that’s why the CYP enzymes and glucoronidation exists. I bet there is some reactive intermediate from depletion of glutathione from glucoronidation pathway saturation.

    I've made this, definitely don't try taking it since a lot of the dosage information is incorrect and it's got a half life of something like 37 DAYS. I wasn't back to normal for nearly a month, my wife still brings it up occasionally...

      I'm gonna take it anyway. What dosage would you reccomend? I was thinking something like 200 mg would be active yet not too overwhelming.

      @@lagrangiankid378 I'd keep it under 300mg which was the medical dose for sleep and I wouldn't take it more than once a month.

      ​@lagrangiankid378 if you aren't going to listen to the SERIOUS AND EMPHATIC warnings to not do this, I would not recommend starting with something so large as 200mg. With a halflife of over a month, you would not process out all of the chlorobutanol for 5-8 months, with 6mg remaining at 5 months and finally reducing to >1mg at 8 months.
      I will not provide a suggested dosing amount as I am not a pharmacist not do I want to encourage anyone to attempt this, but anyone stupid enough to do this needs to be aware that a half-life time is the time to half the original dosage and should plan accordingly. You will be in a highly suggestive state for at least a month, you will have a bad time and you will make regrettable actions.

      @@jacobbrown7367 200 mg is not that large of a dose for something with this level of psychoactivity. For comparison chloral hydrate is dosed at 500 to 1000 mg. The OP of this thread ingested a gram or so if I remeber correctly and this lead to overdosing. And (unlike you) I know precisely what the elimination half-life is (which by the way usually refers to blood levels, not to the total amount in the body). And by the way 37 days is the half-life as measured in vitro at a pH of 7.4, terminal elimination half life in vivo in healthy human subjects is reported to be only 10.3 days (C Tung et al., Biopharm Drug Dispos 1982). I have quite some free time as I'm currently taking a vacation so there's plenty of time to recover and this self experimentation won't have any significant consequence. I know what I'm doing and I'm highly prepared, experienced and equipped. I just asked some anecdotal dosage information as there is little or no human data on this obscure and very old CNS depressant.

      There's a paper out there that even describes terminal self-harm from ingesting mouthwash that contained chlorobutanol. Whether or not the incident was concomitant with other ingested substances due to the past behavior of the poor soul could be debated but it isn't seen as an anti-fungal, antibacterial additive in personal care products anymore.

    For anyone wondering, the effect is literally as having a flu without the pain and fever. You just get enormously letargic and dysphoric and fall asleep very easily. I imagine even hitting your head really hard is more recreational. The taste is like menthol flavoured sand.

      Thank you so much! And yeah it got worse the more I read about it. Interestingly enough in my research I came across what is absolutely the most horrifying chemical I've ever heard of: EA-3167. It has the same effect as datura or extremely high dose Benadryl but with a psychoactive peak of 10 days instead of a few hours..

      @@integral_chemistry Why do I get the feeling that EA-3167 got some research done on it by a certain 3 letter agency here in the states? They had a period where if it caused hallucinations or removed you from reality they figured they might be able to use it as a weapon of war. (Spoiler Alert: Not really, no.)

    You could easily further purify the chlorbutanol through sublimation. Mix it well with some calcium chloride in a beaker to further dehydrate, then heat from the bottom and crystallize on top on the exterior surface of a round bottom flask filled with ice water.

      I like to make every video a surprise that only rewards you if you actually take the time to watch to the end. I think its fun and a nice little way to foster continued interest (jury is still out on whether it'll work lol)
      And yeah I actually noticed the same thing.. I think its a "settling" due to the weight of such a large sep funnel on such a tiny ring stand. I did leave it overnight once or twice and it never failed, but it is still not ideal by any means.
      Think I'll probably bolt some bars to the inside of my fume hood for greater flexibility but also to support some of these heavier objects.

      @@integral_chemistry I made a custom-made lattice inside my fume hood just to accommodate different clamps, I find setting up the clamp and stand every time quite annoying, stands are also prone to tip over which I am not a big fan of.

    I have been a subscriber for several months now and I find your video teaching to be hypnotic. Your intellect and education in chemistry is terrifying to me. Obviously, I know very little about chemistry and synthesis. I know you use your education for good and not evil. It's very easy to see how someone with your background could make things horrible for a lot of society in a very short time. I am in awe of your education and I am thankful for people like you.

I worry often that the pace I go through topics is maybe too fast to the point of being confusing so I love hearing that is "hypnotic" because in a way that's what I'm going for. I love chemistry, and I get so entranced in the process that it sort of becomes this mesmerizing experience where nothing else exists for a little while. I try to channel this vibe through the very focused way I edit down the videos, which I'm very happy to hear you appreciate.
      I also love hearing about your interest in chemistry! Its a subject most people dislike in my experience, so its refreshing to hear people appreciate the wonder of it all.

      The pace is great I wish I would have paid more attention in high school. 40 year pipefitter, no chemistry but, always did love it and was intrigued by it. I have a newfound interest in primer recharging which has fueled the desire to learn some simple synthesis for a few different labs that I will need. I'm in process of gathering equipment now.
      I have some really good chemists to lean on and ask questions to through a couple different groups. You guys are all top notch!

      Chemistry is one of those fields that really weed out the "ill-doers" believe it or not, the mad scientist that media portrays is a stock fiction character and nothing else. People with unmotivated malignant intent are usually addled by extreme emotion and are usually not the types to go through academic or technical education. The worst harm happens by accident, or gross negligence by administrative authorities. This is why you see most instances of mass violence committed with guns or garden variety explosive devices, and rarely with chemical, biological or radiological weapons. Media loves portraying violent anti-social types "the psychopath" as these smart, coldly calculating geniuses that hold dark secret knowledge that they apply flawlessly in the execution of sick fantasy or whatever, but real life criminally insane perps usually have very low cognitive capacity. The more intelligent ones are rarely found in violent crime or terrorism, but are the ones in business making life as a worker in corporate hell. The fear of the knowledgeable/literate dates back to early medieval times and has sadly been deeply embedded in our culture, people fear computer literates too, and respect their mechanic about as much as their dentist.
      Just had to point that out, dont fear the knower, the knower is likely why you survived childhood and enjoy a relatively peaceful existence :)

    So I tried this back in 2017. I ate some, and despite its flavor, didn't puke. It was like plastic cloves amd didn't knock me out at all. It was a waste of chloroform, albeit the science was cool. You did it way better than myself, and you were right to not eat it. Think headache ❤

    Runaway haloforms are NO joke. I was doing one in an erlenmeyer flask and lost control of it. It sprayed hot bleach into my face and managed to get into my eyes, clouding my corneas for the rest of the evening. Instead of going to the ER, I went to sleep. Next morning I woke up fine. That was my last haloform

      Glad to hear you were fine, that sounds terrifying.. You aren't kidding though, the sheer heat these type of reaction generate are honestly shocking and you really have to get it as cold as you possibly can (and even then expect the worst case scenario)

    The smell becoming overwhelming and gross is probably due to how our scent receptors oxidize aromatic compounds, something about them becoming overloaded or the accumulation of side-products of oxidation forming complex molecules with other organics (from your tissues - hence the cancer/degenerative disease risk from many volatiles) that also get processed but register as rancid or acrid due to similarity with rotting meat or spoiled fruit smells.

      you're thinking of sodium pentathol and it doesn't make you tell the truth you can even confess to things that never happened so it's worthless for interrogation,there was once a case where the police went looking for bodies a person claimed they buried in central park NY and there were never any bodies they just implanted the memory.

      These types of hypnotic sedatives would likely render the person a deadhead and rather irritable as well similar to the first generation antihistamine like phenergan - horrible in large doses and even worse in even medium ones.

    I’m sure acetone on its own could be a sedative just based on how nasty it smells. It might end up being a one time use kind of thing but I don’t know. I’m not a chemist.

    As it is only slightly soluable in water, I take it that the historic use was mainly done as either a pill or dissolved in something like alcohol

      I'm assuming a pill to be honest I'm not entirely sure. It still has limited uses these days but the concentration is so low they do just dissolve it

    @Apoptosis: Since this platform is ghosting all answers to my answering comments (accept for the ones from the creator himself, but even my own), I have to answer your question as a new comment:
    Never heard about the interfering properties of ammonium ions on detection of Mg with azo violet, so I tried it out immediately: no blue colorlack was formed nor did any other color change happen (except to yellow if the test solution is too acidic).
    Did you use distilled water to dissolve the ammonium salt? Azo violet is so sensitive on magnesium that it will even detect the traces present in tap water.
    Indeed ammonium often is an interfereing ion in analytics so it has to be boiled off as ammonia with sodium hydroxide beforehand then.
    But in this case it doesn't cause any problems.
    But there's one thing to take into account with this dye: the alkaline violet solution is sensitive to air. It becomes a brown liquid lost its indicator properties when standing open for a longer time. This can be prevented by adding a little ascorbic acid - and preparing the solution freshly everytime when needed.

In any case I hadn't tried the ammonium ion test yet (only read about it on Wikipedia). I also didn't realize until working with it just how EXTREMELY sensitive it is to magnesium ions. It's to the point that when diluted in a sufficient quantity of tap water it is certainly more blue than violet, and when I added 2M magnesium sulfate to a freshly-prepared alkaline sample of azo violet, I got a thick blue precipitate.. which I can't remember whether or not you mentioned in your video.
      The intensity of the colors with this one are actually such that it's hard to get good footage, and in all honesty I might just be using to much of the azo dye. This stuff seems like a little goes quite a long way

      Honestly I'm shocked it took me so long to get around to this one. Very straightforward and satisfying. I'm a big fan of any organic reaction where the products have vastly different properties than the reagents (especially a vastly different smell)

      @@integral_chemistry that’s how I knew I succeeded was the smell. Ofc the chcl3 I used was made from pool bleach and was not distilled before using it for 2nd step. Sample prolly had sodium acetate and KOH impurities even tho I tried to remove KOH via filtration. Chlorobutanol is hard to store it likes to sublimate on the lid of the jar I put it in.

    Idk how accurate this is but I´ve heard that Adolf Von Baeyer synthesized barbituric acid from human urine and apple juice, to make sodium thiopental? Maybe this is something you could try and make a video about. Definitely gives the opportunity for some good video titles; "Making anesthesia from apple juice and pee" 😂

      Huh I've never heard of that before but definitely worth looking into. I know you can make barbituric acid from malonic acid (which does occur in apples) and urea (which is definitely in pee lol). I'll have to check it out and see if its feasible

    I was expecting this to be a synthesis of chloral hydrate. I remember hearing about 20 years ago that it's a very easy synthesis. Requiring you to just bubble chlorine gas through I think ethanol. Which sounds suspect and comes from the same source that claimed you can separate ether from hexane by mixing it with water and draining off the bottom water layer. But it's got me wondering how easy/hard the actual synthesis of chloral hydrate is. It would be a real treat if you could cover this one. If legally possible.

    The concentration of the developer solutions influences the color depth of the dyed textile.
    Here is a recipe:
    Solution A: 10 ml diluted sodium hydroxide solution, a little beta-naphthol. The dyed material is immersed in the solution. Solution B: 25 ml water, some sodium nitrite, some p-nitroaniline, 10 ml diluted sodium hydroxide solution. The solution is cooled to below 5 degrees Celsius and then 25 ml of diluted hydrochloric acid is added until it turns yellow. The dyed textile is then transferred from solution A to solution B.
    The recipe is universally applicable. Depending on the desired dye, the phenol and aniline components can be exchanged accordingly.

    the metabolism of the substance occurs in the liver and the 4 carbon chain might be more fat soluble leading to longer time for total bodily elimination. Just a thought

      Ooh honestly this is one of the more feasible ideas I've heard. It is unfortunate that with chemicals like this that have clearly better alternatives, nobody is ever going to really bother with clinical trials at this point and so we can really only guess at the biochemical mechanisms for such things

    Interesting and thanks. Don't think that I've heard of this one and I agree about the solubility, in fat most likely. Chloralhydrate used to be a very popular item, back in the 50s, I've heard. As it was cheap and easy to make, relatively, anyway. Medical doctors used to write a lot of prescriptions, back in the day. Never made it myself, read about it.

      Hm so maybe I wasn't using too much.. this stuff is just STRONGLY pigmented.
      As a side note I made para red as well and I want to try using it as a fabric dye, but I read that in order for the dye to be permanent it needs to be coupled with phenol or aniline. You know anything about this?

      Yes, azo violet is a fairly rich dye.
      The reference to phenols and amines refers to the coupling process itself and does not indicate an additional work step for fixation.
      I have to admit that the last time I dealt with textile dyeing was in my senior year - that was over 30 years ago.
      So here is a translated text from one of my school encyclopedias at the time:
      Developmental coloration: In contrast to direct dyes, developing dyes are created on the fiber. They are often azo dyes. Two components are required for their production, namely an aromatic as a coupling component and an aromatic diazonium salt. The coupling component is dissolved in a basic, aqueous medium. The fibers, which are mostly cellulose fibers, are soaked with it and then dried. They are then treated with the diazonium salt solution. The coupling reaction that creates the dye therefore takes place on the fiber. Like the vat dyes, the developing dyes are not water-soluble and therefore very wash-fast. Naphtol AS (3-hydroxy-2-naphthoic acid anilide), a derivative of naphthalene, is often used as the coupling component, which is why it is also referred to as naphtol-AS dyes.
      For your coloring with para-red this means that diazotized 4-nitroaniline and β-naphthol would have to be coupled directly on the textile.

      That is definitely possible, honestly probable. I'm convinced there are definitely multiple side reactions going on here, not least because after the formation of the deprotonated version of the chlorobutanol, there a big chance that free oxygen just displaces a chlorine and closes in a ring with the adjacent carbon

      BTW I read that azo violet gives a color change with ammonium similar to the blue color change with magnesium. Would you happen to know anything about that?

      Ammonium often is a disturbing ion in analytics, so it has to be driven out beforehand by boiling the test substance with sodium hydroxide solution.

      I doubt it would be using this route, and I can't find any literature about it. May indeed be possible by another route, but fluoroform is really hard to work with (boils at like -80 degrees) and deprotonation wouldn't work due to the extreme electronegativity of fluorine.

      Hm.. I'll have to ask him about that. Potassium hydroxide is so crazy soluble I didn't consider that a possibility, especially since it has to dissolve to react in the first place. Never know though, definitely worth looking into.

      @@integral_chemistry I think it’s one of those things where, it’s not super soluble in organic solvents, but moreso, it’s driven forward by the higher solubility of the product relative to the hydroxide. Also, it would make a lot more sense as you saw in your mechanism. No formation of carbines or loss of product due to some weird side reaction. I suppose also, you could filter it, dry it out and see if it’s hygroscopic, or reacts with something like calcium to make calcium hydroxide, which wouldn’t be the case with potassium chloride.

      @@toseltreps1101 I really don’t think that could be it just by how much there is. If you look at the equation, hydroxide is regenerated, so it would make sense that’s what it is. Also, in basic medium, conversion of KOH to K2CO3 doesn’t usually happen that readily and that quickly

      @toseltreps1101 also, as I stated, you could easily determine if it was potassium chloride by washing it well, drying it, redisolving it in water and adding calcium chloride. That would precipitate calcium hydroxide if it was just powdered hydroxide, but nothing would happen if it was potassium chloride.
