That Luke, her patient, sweet, reliable, will-put-up-with-her-no-matter-what Luke could ever be her ex boyfriend. This scene is so sad, but I love the acting from both of them.
I think that’s why she felt so Guilty, with how much he put up, she may have taken him for granted, and she didn’t realize that until he finally gave up, considering how much he smiled through, she realized she must have completely hurt him for him to walk out, even then he walks out in a complete manner, and not leading her on, if he felt compromised, he walked out without compromising her possibly in the future, in a past scene she has a dream and is mad at herself for not telling him anything and playing it cool when he said he was all in, you can tell how guilty she felt for telling him she’s all in a grocery store after he was on the verge of breaking up, basically too late, and throughout their relationship he was open about his feelings, showing his commitment, and she couldn’t even tell him the truth about a harmless night spent with Chris

His "okay" is the same "okay" that he gives after she proposes to him and says she just wants to be happy. Just letting her lead. If she had said, "Luke, I want you, please take me back," in this moment, he would have agreed instantly. He had enough time and he was ready. But instead, she told him he was right and it was over. So he went.
At 3:30, there’s a change in Luke’s body language and as an actor I love how that was executed. I may be reading into this, but that’s the manor he takes when he’s regretful or hurting and trying to cover it up. it’s been present in a lot of other episodes, mostly when Lorelai was with another guy or when someone was talking to Luke about her, never in all the time they were together (until Christopher, but that’s another rant). To me, it’s the change in his mind. He feels the shift in her tone. Before this his body language was the same as when they were a couple, meaning it had t really sunk in that they were broken up. Now he realizes she’s not gonna cry into his shoulder and love him again just like that. He realizes he screwed it up, that they’re done. You can see it in his face, how he raises his shoulders and tenses up his head and neck, which is usually (in animals, at least) a defensive gesture, trying to show strength and hiding any weakness (no relaxed muscles, tensed jawline, etc.). This reinforces my theory that he’s realizing it’s over between them, and he’s trying to act like he’s okay, trying to hide his pain and going back in his shell. Even if he doesn’t realize he’s doing it, it’s there. It’s the same stance he takes with her the rest of the time they’re broken up. This may just be me over analyzing but idk. He kept trying to get her to come to the play to work on the costumes while he worked on the set itself. It seems like Lorelai finally accepts the break-up and Luke realizes he's lost her during that one scene (i.e., "Do you love me?"). From then on (i.e., in So Good Talk), Luke just goes downhill, and Lorelai's working on moving forward. But your points about Luke frowning, hunching over and seeming as though he's 'screwed up' signify that he's subconsciously aware here. So his purposeful steps to try and get them in the same area (i.e., Jews/Chinese food) are because he's lost control which happens here, not later (as I originally thought). Not quite a Ted talk here, just my ramblings. 😉

Luke shows many signs of regret when he and Lorelai were broken up. Like his face at 4:15. You could see that he was worried😭😭❤️❤️
