I found that there are a lot of advantages to using a backpack for touring/bike packing. For one it's cheaper than buying all the racks, paniers etc, you can bring your stuff with you when you go inside a store on your trip, it's a lot easier to lift your bike over a fence or gate and my favorite is if you're wild camping and you find the perfect spot to camp but there's a really steep embankment to go up or down to get to the spot, you can stash your bike then walk to the spot with your gear on your back instead of lugging 60+ awkward pounds up or down a sketchy hillside
One other thing is that the straps hold the sweat against the skin and don't let it dry. Last time i used a backpack for an all day ride I had a rash/acne everywhere the straps touched. And that was with a good pack that distributed weight well
I had no issue using a 25 liter Vaude backpack during a recent 7 day roadbike trip in the Ardennes. Weight was about 6 kilo and I paid special attention to weight distribution. Other than a small saddle bag I had no other bags on my bike

In 2 weeks I am cycling across Thailand - 800km, with a backpack only, oh and flipflops, its 35 degrees and I am ginger... but.. I am stupid :) so, it is what it is.
I would love to build a bike I can pack/camp with for a few days!

Man I have a Teton sports hiking pack I take with me anytime I go bike packing got fork bags frambags too let's me carry so much more stuff and the backpack I've never had problems with but I am on a ebike with a plush bench seat lolz 😂
Some backpacks do have pads on them that will give you cushion
