A little bit of additional context is what happened just before this. I'm not sure exactly what since I didn't watch Subaru's stream but Rushia's, but Subaru and Flare were doing something together in Minecraft, and then in chat when they finished Subaru thanked Flare and then wrote "I like/love you!!!!" ("sukiiii!!!!") not just once but several times. Rushia, who likes Flare, saw this and it was the reason she decided to play a prank on Subaru to begin with. She still didn't intend to throw the poison potion but instead just forgive Subaru with a little prank, but Subaru on top of the chat messages of like/love to Flare also mistaking the one pranking her as Flare was what lead to Rushia deciding to throw the poison on her.
