Thank you - as others have commented, you nailed it. I have been madly doom scrolling everything accessible regarding ALS on the web, and my ChatGPT research has been crazy helpful. With a lot of research only since the beginning of September, I reached out to and was admitted to the Johns Hopkins ALS Clinic. I literally do not know at this moment what’s going on, but they admitted me. I believe the fact that my mother succumbed to ALS at 85, and one of my first cousins was just recently diagnosed. I have mess of symptoms and medical history, which I can trace to everything but ALS. Yet, I go in on Monday to find out what the diagnosis is. I couldn’t be more prepared to advocate for myself, to articulate a solid history of my medical history, with records… not that I am going to beat it, but I sure will meet it. I have hope that somehow this isn’t ALS. Will come back when I know more. Thank you all.

It was good and useful. But I think you had a tiny mistake in distinguishing upper motor neurons from lower motor neurons. You said "lower motor neurons in brainstem and spinal cord", while all neurons in these areas are upper motor neurons. Thanks though, it was pretty useful and clear❤
Are the mechanisms of 1) glutamate-mediated excitoxicity and 2) mutant protein aggregation to prions both happening in ALS, or is it one or the another that causes 3) inflammation/release of damaging chemicals?
Sadly there are important parts missing in this picture, especially the toxic effects of dietary sugar/fructose on brain metabolism and the effects of insulin resistance on the neurotrasmitter-balance...
I don't suppose you have rough estimates of percentages of apoptosis vs. ferroptosis cell deaths. I haven't seen any such numbers yet in published studies. Also curious about the noxious chemicals- are these cytokines, reactive oxygen species, reactive nitrogen species, or something else?
@@Leonardo-cu7fd I hadn't heard of cytokines referred to as 'noxious chemicals' before. Some are useful in chemotaxis, but in this instance they are harmful. The glutamate at the NMJ is problematic. About 4% of what shoots across isn't acetylcholine, but calcium. Accumulation causes other problems. There's a LOT going on in ALS that would take many videos to explain. Thanks.

My young athletic son died of MND. He was very fit and had been using a muscle building shake to build body muscle. I am convinced that his illness was caused by Maximuscle I have tried to have a dialogue with Glaxo Smith Kline regarding this but they have dismissed my concerns. The product is high Glutamate and high protein and I think his body couldn’t recycle sufficiently. I am not a scientist just a heart broken mother. Could you help my cause please. You're right, he died because of the supplement, this disease is common in athletes, Lou Gehrig died the same way, by using anabolics steroids, the explanation of the etiology of ALS in this video is wrong, is not the glutamate is LDL (bad cholesterol), the whole explanation is long, so I cannot delve on the topic in this comment.

Due to having shit memory when I’ve always had a sharp memory which has lasted for the last 2 years getting worse, muscle twitching which only started in some body parts now it’s all over and in my tongue, noticing muscle weakness & loss in one leg and one hand, always tired asf! Feels like I’m slurring when I speak for too long, overall muscle fatigue and my postures getting shit sloooowly. Tried to see a Dr about it but here in Nz they’re trash and what’s funny is Als/Mnd has the highest rate here in Nz for frick knows why?

@@marika_meek doesn't work that way you would notice weakness loooong before any loss of muscle. Weakness with twitching but weakness is the king the num 1 the head honcho the main squeeze

Microglial cells release inflammatory cytokines.

there's no relationship, the protein aggregation and amyloidosis is key in ALS but the glutamate theory is bullshit.
