It should be a Stephen Strange Variant who left Ancient One after figuring out that Mystic Arts cannot fix his hands. Instead participating in Deadpool Experiment turning him into Strange Pool, played by Benedict Cumberbatch himself.
I would love if Doctor Strange is in the movie, it would definitely be one of the reasons I'm going to see Deadpool & Wolverine. It seem like he would be since the Portal scene is in the trailer.

And many reports swore that Deadpool was going to appear in DS MOM -- even going so far as to say that you could see Deadpool in one of the movie posters. But he wasn't. If Strange appears, I'll be happy, but I'm not holding my breath.
While Deadpool & Wolverine plot specifics are still largely under wraps, it seems unlikely that Loki will be essential prior viewing. The former God of Mischief will likely have an important role to play in the future of the MCU, but it seems unlikely that Loki's new role as the isolated protector of the multiverse will be significantly advanced here (if at all). Tone matters, too, and it's easier to hand wave additions like the TVA in a comedy known for non-sequiturs than a more "serious" story like Doctor Strange in the Multiverse of Madness.

That's denfently Doctor Strange to appeared in Deadpool 3 because this month Deadpool & Wolverine is reshooting to added more cameos from the Fox X-Men Universe and MCU Heroes by the End of the film.
