Yes, when set up properly, they can protect against DoS attacks regardless of whether it's done at a specific service level or at the API gateway level.
Most cloud API gateways (ex- Azure and AWS) provide easy to set up rate limiting options so a lot of development teams tend to go for this option. Plus setting it at the entry point (like your API gateway) means less services are exposed to any attacks.
