Sir, In shaded region points should be selected as per your need. If you need higher amount of oil to load maximum drug than select point with maximum oil. If you need to focus globule size and stability then select point with higher amt of surfactants. You may also employ DoE for selected region.

during preparation we use in % or in ml? if in ml, should we fixed the total surfactant concentration right?

1) Theoretically, it should be just before turbidity appear.
2) First prepare Smix (so three phase: Oil, SMix and Water) for pseudo ternary graph
