I really believe they are soul mates. I think they are different things to each other every lifetime. They were probably mother/daughter, husband/wife, siblings, best friends, rival monarchs, etc etc.

The first Sam death still makes me cry every time. Jensen just knocks it out of the park.
The hug at 9:39, it's more than brother's hug. It's like comforting motherly emotional hug. Sams expression says it too.
The only sad thing is that they only hug when something bad happened.
