It's not confirmed that All for One is Midoriya's father, however there's a possibility because it would make sense for All for One to have children with women across the country and take their quirks. All for One has the ability to take away their quirks before they even get them and it's also possible for Doctor Garaki (All for One's helper and the one who creates nomus) to take role of the family's doctor and confirm that the child is quirkless. As you can see in the manga AND the anime, the doctor that diagnosed Izuku looks exactly like Doctor Garaki. BUT! there's still a possibility of Midoriya and Shigaraki being blood related cousins. As we know, Shigaraki's grandmother was a hero, Nana Shimura. And when we look at her, she almost looks identical to Midoriya's mom, Inko. The hairstyle, the quirks, EVERYTHING on Inko's character just screams Nana. Yeah the last names are different but let's not forget the fact that Inko was/is married. And if Nana is really Inko's mother, that makes her Midoriya's grandmother which would make him and Shigaraki cousins.
