Can we change field weight in one or more fields to reduce doses in such points?

You can change the relative weighting of each field after optimization. However that is something that is normally only done with 3D fields. That is because the effect on the total dose is not predictable due to the complicated fluence of each field.

how much time it takes to complete contouring of one case?

Can depend. Usually around an hour but could be longer for more complicated cases. With new autocontouring tools, it could be much faster.
