I dont think they could find the chamber even if they knew it was there I think it was under the fidelius charm and Harry only found it because he is part Voldemort

Ron and Lockhart accompanied Harry to the Chamber and, we have seen in Order of the Phoenix, that without reading the note from Dumbledore, even Harry could enter Number 12, Grimmauld Place. I think it is less likely that there was a Fidelius Charm in Place.
