Bacteriophage used in genetic engineering as a Cloning vector. It helps to transfer desired gene into the host cell and also helps in replication of gene of interest.
Using bacteriophage we can obtain specific production like human insulin hormone.

As we cannot see phage or viruses through naked eyes or even light microscope then how can we insert gene of interest into phage DNA. Maybe gene of interest will larger than whole DNA of phage????
We can see phage through compound microscope and the whole gene of interest is not inserted in phages only it's small fragment is inserted ☺😊

Sir we know that when any virus dna enters into bacteria then restiction endonuclease cut this dna so why restriction endonuclease doesnot cut the bacteriophage dna when it enters as a vector???? Pls sir answer this question
Because the phage DNA has an adaptation to change its chemical e.g its nitrogenous bases undergo methylation (addition of CH3 grp) so enzyme being strict to shape and structure of substrate can't now act on it

Sir the lecture was great👍 but I have a question😅... When the Bacteriophage will insert Recominant DNA in bacteria, then this Recombinant DNA will combine with bacterial DNA and phages will form which contain all the genes including gene of interest.....Right , then how we will extract gene of interest from bacteriophage????😓😓
We again do not need to extract the gene of interest but if we want to extract then a specific enzyme called restriction endonuclease is used and is putted on the gene of interest at specific sites and cuts it and hence we can extract the gene of interest.
And recombinant dna do not combine with bacterial DNA it destroy the bacterial dna and takes over its machinery
