Yes you can. It depends on the tire/rim combo. If the fit is snug enough, and a tire with a quality bead, a few quick pumps will pop and seal the bead. I've found the GP5000's easiest to do with a hand pump.
In 14 years I've never used anything but a hand pump. If you have problems getting the air into the tire, make sure the tire is spread on the rim as much as possible and squeeze the tire down over the valve at the valve with 1 hand, and pump with the other. It will get the air started and then you can just pump with the hand pump with both hands.

Yes. Schwalbe is a bit tough but doable. I apply liquid soap in the beads. Continental 5K STR is even faster to seat with just floor pump.
