    Hey! Small thing I would like to point out. The rflp fragments you show on the sketch are all equal in length where you mark the RE sites. Wont separate on gel. :) Also the smaller three fragments would migrate much faster than the two large ones.

    Sir for RFLP do we extract whole DNA of a cell (6+ billion bp) and restrict it with a restriction enzyme? Or do we extract only a part of the entire DNA and then restrict it? If so how do we do it

    which type of nuclease enzyme is best to use for degrading all DNA in a sample??
    plz explain
    and one more
    you want to degrade all DNA in a sample to avoid contamination..will you use exonuclease or endonuclease and what will be the reason for selection??

    I have a doubt. In AFLP, if we already know about the restriction enzyme sequence flaking the sequence of interst, why do we need adaptors. We can simply use primers against those restriction sequences. Please do reply

    I have a querry,...is it not that we don't require sequence information in AFLP too..just the adapter sequence is sufficient

      May be, it should be known to select the particular Restriction Enzyme , which do not cleave our Target DNA. I don't know,it's just a guess.

    Hi sir you have tell about distinguish between these three techniques but this is not enough it requires more emphasis on how three techniques are different, this explanation seems like these three techniques are useful for same purpose, you should have explain actually what is the main purpose of these 3 techniques and how they exactly distinguish this is important thing because
    In molecular biology techniques some times seems all techniques are useful for same purpose that's why I am emphasizing on purpose & distinguish
