Amazing video!! I have a question, so in the LoxP mouse, all the tissues will have the gene you want to delete flanked by LoxP sites.
And only in tissues where Cre recombinase is active will you have the gene deletion occurring.
Do the LoxP sites around the gene interfere with the correct transcription/translation of the gene in tissues where Cre recombinase is not expressed? I am wondering whether the LoxP sites could interfere with the translation of the gene.
Good question, in most of the cases it does not interfere. The loxP sites are small.generally they flank exon and not in orf.

You need to use the cre ER system . Tamoxifen inducible Cre-estrogen receptor (ER) fusion protein. For this, Cre is fused with the ligand-binding domain of a mutated ligand-binding domain of ER, which does not bind to estrogen but binds to tamoxifen with high affinity. Cre-ER proteins are sequestered in the cytoplasm via association with the HSP90 chaperone. Upon addition of tamoxifen, tamoxifen-bound Cre-ER dissociates from HSP90, translocates into the nucleus, and carries out site-specific recombination between flanking

Hi, can the cre recombinase travel to other tissues (even though it has a tissue-specific promoter) and knock out the GOI e.g. the cre is brain-specific but can it travel to the liver and knockout loxp flanked gene of interest? And is the loxp flanking also tissue-specific?

In order to study Dopaminergic Neurons morphology, why not adding a GFP reporter gene directly after the specific promoter ? Rather than Cre Gene and a Flux stop GFP gene.
It's also possible, in that case that mouse line has to be generated

Cre is an enzyme ( it's a recombinase enzyme). Cre loxp system can be used to knockout genes.
