  • As of March 17, there are over 180.000 people infected worldwide by the new coronavirus SARS-CoV-2, which can cause the coronavirus respiratory disease COVID-19. Recently the WHO named the outbreak a pandemic and now globally cases are rising sharply, outstripping the Chinese cases.
    As the outbreak of the disease is reaching a high number of countries all over the world, the general public awareness and scientific interest are also on the rise. Currently, the virus virulence, how it is transmitted between humans and the possible treatments of the disease are being actively studied. As there is already a lot of information available, we have here gathered the known information on the molecular organization and structure of the virus, thus creating a dynamic and scientific accurate 3D animation of SARS-CoV-2!

    Many different drugs and therapies are being trialled and used on patients with Covid-19 and rapid progress has been made not only with vaccine developments. There are some positive results, which may be beginning to bring the hospital death toll down, but there is still a long way to go towards something a cure all. In our video we mention Remdesivir and convalescent plasma, and research has shown they are working, but here are 4 others, which have received some attention recently, we did not kow about when our video on potential COVID-19 treatments was made.     1. Dexamethasone
    This is the success story - a very cheap drug, in plentiful supply around the world - that has been shown to save lives. The low-dose steroid was responsible for the survival of one in eight patients on ventilators during the Oxford University-based Recovery trial. The results were announced in mid-June and the steroid is now standard treatment for the sickest patients. The data is reliable because it comes from the biggest randomised trial of Covid-19 treatments in the world, involving almost all hospitals in the UK. Dexamethasone has been around for 60 years, so is out of patent and cheap.
    2. Tocilizumab
    A number of anti-inflammatory treatments are being tested to see if they can help. When the immune system responds to the coronavirus, it can cause inflammation. Tocilizumab, an antibody normally used to treat rheumatoid arthritis, is given by injection to block the inflammatory protein IL-6. Patients given the drug during the serious Covid-19 outbreak in Italy in March appeared to benefit and were less likely to end up on a ventilator or die. But the numbers were relatively small and this was an observational study, not a randomised trial set up to judge the outcomes of people with and without the drug. The Recovery trial is also investigating Tocilizumab. Roche has said that recent trials have been disappointing, but the company wants to explore other ways of using the drug.
    3. Blood pressure pills
    Scientists think people on medication because of their blood pressure would be well advised to stay on it. A paper from the University of East Anglia found that the risk of critical illness or death from Covid for people with high blood pressure was found to be significantly lower if they were taking angiotensin-converting enzyme inhibitors (ACEi) or angiotensin receptor blockers (ARB). That does not mean those drugs would be helpful to anyone with coronavirus infection who does not suffer from high blood pressure.
    4. Inhaled interferon beta
    A small study from scientists at Southampton University suggests that an inhaler containing the drug interferon beta, used to treat multiple sclerosis, could be effective. The three academics behind it, who formed a spin-off company for their product in 2004, became paper millionaires when they announced the results of their trial on Covid-19 patients in July. The trial involved just 101 patients at 9 hospitals. Those inhaling the drug left hospital sooner and were more likely to recover, but the data was not published in full. Larger trials are needed.

    The outbrake is sometimes we feel is due to deforestation and urbanization - due to which the viruses of animals are becoming viruses of humans as well because the gap between wildlife and humans is vanishing.
    Sometimes I think Humans are also like a virus to earth :-

    *difficult to destroy the over population (due to machines)

      Homo sapiens, humans, are animals. These sort of viruses are zoonosis, i.e. they were initially in other animals, but now infects other species as well. In Wuhan this happened, because on a fish market they store different animals in the same cage and/or close by each other, thus the chance of viruses to become zoonosis is increased. This is not exactly because of deforestation and/or urbanization.

      Yes you are right, it is not directly related to deforestation but The exploitation of wild life is. But again as Bill Gates said More of these Virus Jumps can happen in near future. So indirectly The deforestation, defying the idea of co-existance, Not respecting wild animals and their space in earth would lead to other pandemics as well.     We got sick in January 10th and then my son on the 18th in Costa Rica.
    I asked my health professional what was happening and he said it was a tropical flu.
    Symptoms
    Head ache 3 days before symptoms appeared.
    Bone breaker 1st
    Fever that night.
    3 days of pain and discomfort.
    Very dry Cough lasting 3 weeks.
    3 am coughing attacks when laying down on our backs.
    Dry cough, like saw dust stuck in our lungs. Nothing would come up.
    It was painful.
    My son 14 years old. I'm a smoker and 54 years old.
    Exact same symptoms exact same cough.
    Lasted about the same amount of time for us to recover.
    It did notable damage to both our lungs as we had trouble breathing and doing heavy cardio afterwards.
    We had morning cough like smokers cough for weeks after we recovered.
    After 2 months we are both good and respitory system back to good working order.
    What did we have?
    I can't get a straight answer.

    Dear Biolution, I congratulate you for the impressive animation. I would like you to suggest to prepare a new animation. Because there is a great need for this. As you might know, SARS-CoV-2 needs two protein from host cell: ACE2 and TMSSR2. ACE2 is angiotensin converting enzyme 2 and TMPRSS2 is transmembrane serine protease. It would be very nice if you can prepare an animation related to binding of spike protein of SARS-CoV-2 to ACE2. As you also might know that this is not enough for virus to show its effect on host. Other protein, TMPRSS2 is also needed for priming the spike protein of SARS-CoV-2. I think this could be animated and it is needed.       This would be quite a project, we considered to show the cleavage of spike by ACE2 and looked at the structures. Not so simple! We'd gladly do it if we can find someone to pay for this work, but for TMSSR2 not even a structure is identified yet.

    Not ACE2 but TMPRSS2 protease cleaves the S protein. ACE2 is the primary receptor.

    I believe there are options for prevention and treatment that entail two possible approaches: We know the primary avenue of attack are the lungs, and once in, they attach by means of the spicules or "spikes". Their primary target or "receptor" are the ACE-2 or angiotensin-converting enzymes, same as that of SARS-CoV, on the epithelial cells of the lungs. Right there is one strategy for a form of intervention using nebulized or detached bonding receptors, akin to chelators or in fact serpins, serine protease inhibitors.
    Beside Remdesivir, Pyridone type inhibiting compound 13b will likely emerge as promising along these lines within the body. This compound disables the sars-cov-2 cleavage enzyme on the attachment spikes. Since this is a papain-like enzyme, the inhibitor will not affect the human serine enzymes, like trypsin, amylase, etc.
    It could be used externally as well to attach aerosolized virii and they'll be inactivated even if they are inhaled.
    Another approach is in terms of direct digestion of the virus by hydrolases like amylase, trypsin, chymotrypsin and lipase. Intramuscular injections of amylase and trypsin were proven to resolve malaria very effectively by Lambelle in the early part of the 20th c. The ratio of amylase to trypsin was 2:1, and was well tolerated. His paper is available as a pdf. While malaria is not a virus, the process of infectant inhibition of native enzymes taxing the whole system is akin and the strategy of bolstering it is rational.
    It is worthwhile to also research the potential of external nebulized colloidal silver in air systems and bound to fiber masks, as an effective means of external denaturing the virus. As for inhaled nebulized colloidals, I am not aware of its possible utility or risk one way or the other.

      It was isolated in 2019 and this determines the name. This makes sense as in the past without advanced sequencing molecular definition was laboursome and lengthy. Calling it after the number of infections makes little sense, as this is something we only know later.

      Chloroquine, like other drugs (ie remdesivir) potentially inhibit replication of the virus in the cells. Some people think chloroquine works by altering the pH in the lysosomes, but the mechanism is unknown. It is not presumed to bind the virus.
      Providing soluble ACE2 could prevent entry of the virus.

    When the Spanish flu killed an estimated 50 million people , nurses found out that by taking patients outside when the temperature was warmer , patients got better . This telling me that the virus needs colder temperature to survive ...

      Soap is sufficient to inactivate the virus. Look in our coronavirus playlist for an explanatory vid. If infected people wear simple masks, it prevents from spreading the virus in little droplets.

      there are approaches aimed at this, antibodies from convalescent patients would most likely bind to spike proteins and prevent interaction. Another approach would be providing soluble receptor ACE2

      Also the other coronaviruses can cause severe respiratory tract infections, but it is much rarer. Somehow the SARS, MERS and SARS-CoV-2 appear to evade the immune system more eficiently. Their virulence may be to do with the receptor ACE2, which serves as a protease in angiotensin signaling. This appears to impede the response to the virus further. The more benign coronavirsues do not use ACE2.

It is quite a coincidence that the Ace2 Receptor produces enzymes just to control blood pressure. I would very much like an infectious disease specialist to study the amount of ACE2 "ECA" in the cells of infected people or those who use drugs to control blood pressure.       Bacteriophages work well against bacterial infections. But there is no virus to directly attack another virus, that has to be done with a retrovirus that infects a number of healthy cells, and then hijacks them to produce antibodies for the actual virus ravaging the body. I am not a virologist, so I might not be 100% right on this.

      @ajith0306 Well, just knowing what it looks like (and this is just an artist's impression using the known protein structures of the surface proteins) will not help you to diagnose with a PCR test.
      Viruses are so small, you need an electron microscope to see them.
      A PCR test is fast and reliable.

      Most viruses of higher eukaryotes do not show a strict separation between lytic and lysogenic cycles, as they lack the capacity to incorporate into the host genome. SARS-CoV-2 has not been reported to integrate and therefore cannot follow a "lysogenic cycle".

      @@biolution Thank you. I see. Regarding the current vaccines, I was concerned about how mRNA vaccines are able to be accepted into the cells without being degraded by ssRNA or miRNA complexes. Do you have any idea or thoughts on that? thanks

    Uuuuuhm SARS-CoV-2 does NOT have the hemagglutinin-esterase protein. Also, the M membrane glycoprotein is missing in this model. How dare you call this a cientifically accurate animation? :p
