Actually one of the key things about the whole Rocky Horror experience, is the audience participation. You don't just pay money to see the show, you pay to be part of it, through the participation. RHPS isn't exactly a conventional show, you don't go in sit down, listen and leave, you dress up, don a wig, head in, have a couple drinks, and watch an amazing show that you can participate in from the audience making the whole experience much more fun for everyone :P
Now THAT is how you acknowledge the callbacks without breaking the scene. The best fans know which callbacks work with the live show. The best actors know when they're coming.
As much of a Rocky Horror purist I am, and as much as I really dislike a lot about this production, I think Terrence Mann is actually amazing as Frank.
The Rocky Horror Picture Show played in a local theater nightly. In 1981, I declined the chance to attend a viewing with audience participation while the film played. I have kicked myself ever since for turning the invitation down.
The audience usually yells, "It's a palindrome, a**hole!" in response to Brad there. Frank's line isn't an anagram, it's sort of a line-unit palindrome. However you interpret his meaning it up to you....
Seriously though, can anyone just imagine what would’ve happened if Terrence Mann would’ve brought back Frank in 2000? I guarantee you he would’ve been won the Tony instead of Nathan Lane

Am I the only person who thinks the audience participation is EXTREMELY annoying? I hope I don't sound like a grouch, but you pay good money to see this awesome show, not to hear everyone else yelling at the stage

The joke is that it really isn't an anagram, it's a kind of palindrome.
It is in no way a palindrome, but the term anagram fits "unlockamind" "unmindalock" quite perfectly.
