Great vid. It falls in quite nicely for me as more relaxing than "hardcore" science explanations or reading actual papers, but also not too meme-fied and lose all seriousness. Just the right amount of information and jokes imo.
tiniest detail that I noticed was incorrect: naloxone has a hydroxyl group on carbon-14 (the carbon where the chiral hydrogen is highlighted) instead of a hydrogen. this is because without the hydroxyl group, most allylmorphines also function as k-opioid receptor agonists, which can cause problems with confusion, dysphoria and hallucinations.

I saw the drug structure on the thumbnail and my immediate thought was this wasn't cubane on cocaine but an opiate with cubane. I was not disappointed when I found they just added it to all kinds of drugs.
love the cubanized benzocaine version :) I think cubocaine would have much longer T1/2 in the body, so sucking a cubocaine / rafathricin tablet for sore throat would provide much longer local anaesthesia than its currently used benzocaine counterpart :)

to be honest, cyclopropane isn't all that stable -- it was way too prone to produce a BIG BADABOOM in the operating room 🙂it was even more problematic in that regard than ether vapour or ethylene (can be compared to acetylene though) 🙂 this was the main reason why it was phased out as an anaesthetic other than BADABOOM, nausea and vomiting and adrenaline cardiosensibilization -- it was great in the sense that it, much like ether, had low toxicity, wide therapeutic margin, could be used in hemodynamically unstable, highly traumatized, volume depleted patients it also had the ability to provide EXCELLENT analgesia for looong hours after the surgery, much like ether vapor -- which greatly reduced the amount of opioids needed for postoperative analgesia
What amazes me the most is they had no idea if it could be made or if it was stable. At least with complex natural product synthesis you know that it is possible to make somehow, even if that’s through biosynthesis.

This is the English pronunciation which is different to the American pronunciation. “Amide” is also another chemistry word which is pronounced different.

Could this perhaps be used to prevent the metabolism of dextromethorphan? DXM's most medically important uses, as what is basically a superdrug at this point, all share a similarity in that its metabolism needs to be inhibited. Specifically, dextrorphan (DXO) is typically unwanted besides niche uses for dissociative-anesthetic therapy akin to ketamine therapy. However, the other metabolites of DXM are wanted as they are neuroprotective and seem to help cancel out some of the possible negatives that DXM would otherwise have so perhaps it would be foolish to assume a cubane version of DXM would be able to have the same therapeutic success. However, it would likely limit abuse. Another interesting drug that could benefit from it is salvinorin A which has poor absorption due to its fast metabolism. It could help with making salvinorin analogs for use as a k-opioid receptor mediated anesthetic that are long lasting.
i wasn't aware of the race of youtubers, do like Sams channel but didn't see his method. did however watch most of Tom's synthesis and considering his location both his lab/shed in the rural Australian outback idk but seems there was a slight advantage
