I fully believe that their hand holding scene was meant to symbolize Haruka and Michiru having sex for the first time. They're in love with each other, but choose not to be in a relationship the whole season because they feel that will complicate their mission. Due to the enormity of what they're going to do, they give in for one night, but that's enough for their feelings to compromise the mission afterall, since Uranus immediately tries to save Neptune after she is captured by Eudial. Compare this to episode 98, when Uranus leaves Neptune for dead.
I thought it was simpler: Michiru says almost literally that she loves Haruka. The facial expressions in the episode confirm it quite well: Haruka seems pleasantly touched and astonished. Michiru seems reassuring and warm. The gestures confirm the reciprocity of the feelings.
