The thyroid hormone synthesis story starts from para ventricular nucleus located in hypothalamus. Para ventricular nucleus release thyrotropin release hormone which act on thyrotropes kind of cells located in anterior pituitary gland. Then this gland will release thyroid stimulating hormone and it go and work on G protein coupled receptor located on thyroid follicular cells and make special kind of protein kinases and ultimately will synthesize thyroglobulin and sent to the thyroglobulin in follicular lumen. Tyrosine amino acids are residues of thyroglobulin. Iodide will come from diet and salts which will be absorbed from GI into portal circulation and go to general circulation. Iodide will be transported to follicular cells via sodium iodide secondary symport. And iodide will go to lumen from thyroid follicular cells via Pendrin. And thyroperoxidase enzyme plays very important role in iodination and coupling and arrangement of iodine on tyrosine residues of thyroglobulin. This enzyme also converts iodide in to iodine. Any mutation in this enzyme can alter thyroid hormone production. we have big chunk of thyroglobulin with MIT and DIT then it will be transported to follicular cells via endocytosis process and where lysosomes will cut and arrange thyroglobulin in to T3 and T4. And T3 and T4 will work on heart, liver, skin, neurons , muscles and bones. Basically it increase the gene expression of beta adrenergic receptors, certain enzymes involved in metabolic pathways. And most importantly it makes a lot of sodium potassium pumps. It is a big topic. I can’t write here.
