Chiyo is Ayumu's closest friend. Chiyo means more to her, than any of the others do. And Chiyo is the one who cares most about Ayumu, and Ayumu notices this and therefore appreciates and values Chiyo highly and trusts her the most. This is quite obvious if you pay attention throughout Azumanga to the interactions they have with each other and listen to what Chiyo says to her and the thoughts we hear Chiyo have about Ayumu.
So she likely spoke to Chiyo about jumping off because she wanted Chiyo to know her better, and to deepen their bond. And she felt comfortable in that setting together with Chiyo. She may've wanted Chiyo to know for quite some time, but she couldn't just walk up to Chiyo at any moment and bluntly say _"I'm not as happy as I may seem, I'm really not okay, I often feel bad and sometimes I think about killing myself, please help me"_ so when that situation (them standing there together near the roof edge and Chiyo having already initiated the conversation) on the roof presented itself to her, then she acted quickly and made use of that situation, to voice her thoughts to Chiyo that way. Because she felt comfortable in that moment, and she could word it the way she did -- instead of being direct and blunt about it, which would've felt more difficult. Much easier for her to latch on to what Chiyo said and respond light-heartedly with _"Yeah, it's nice... feels like one could just fly away if one jumped off from here..."_ and hope Chiyo would understand.
So yes I think it was intentional. Just not planned: before they got to the roof, she didn't know she would be speaking to Chiyo about that there and then. For some time already she'd wanted to open up more to Chiyo, and simply finally found a suitable moment and suitable way of doing it.
But Chiyo was very perceptive and had already noticed Ayumu wasn't feeling well. Chiyo had already suspected Ayumu might often be awake during the nights, that that's the reason she sleeps in class -- we see Chiyo thinking about that in the same episode as this video is from (Ep 12), but she undoubtably thought about it earlier as well. Surely she also logically wondered _why_ Ayumu would be awake during the nights and connected some dots. And two episodes prior to the one this video is from, in Ep 10, Chiyo sees Ayumu being sad, and later sees her very sad and crying, because she thinks she'll be separated from the others (when the classes were shuffled as they became 2nd year students), and Chiyo is being supportive. And crucially it should be noted that before Ayumu even says anything about that to Chiyo, Chiyo had already noticed that Ayumu was feeling bad and went back to the classroom (where she "knew" Ayumu still was) to cheer her up and talk with her. So Chiyo had long ago perceived a lot about Ayumu. And perhaps Ayumu sensed that Chiyo was perceptive and understanding, which could be a factor in why she felt so drawn to Chiyo and comfortable with her and so on. But Chiyo was more perceptive than Ayumu realized.

Chiyo is so precious, as is Osaka. I feel Osaka on a deeper level. Can't pin down exactly how, because I'm not suicidal, but I do have strange intrusive thoughts like she does. I'm glad she has friends like Chiyo, though.

I thought Osaka looked at the ladder because she thought her lunch would taste better up the ladder. And the other comment is a reference to Chiyo chan's ability to fly. I don't think there is a deep depression message, I think Osaka is on brand here.
When Osaka looks up there, yes, that's because of that (the food). But the preceding part isn't about Chiyo. She's talking about herself, as in "If I jumped off here" or "If one jumped off here". She's not saying Chiyo could jump off. And Chiyo specifically says she's worried Osaka might do that. So it's very specifically about Osaka, not Chiyo.
There's not any "deep depression message" anywhere in Azumanga and Azumanga is meant to be primarily funny and light. But if you look past that, if you look past the surface, past the sillyness etc, you will see that Osaka is indeed depressed and and has suicidal thoughts. It's very subtle and only hinted at (in various ways), but it's there. If you read/watch the whole of Azumanga with this constantly in mind then you should be able to notice it (with the anime it helps if you watch it without sound so you're not influenced by the cheerful music and voices). The two most obvious things that can be pointed out is the first roof scene when Osaka talks about jumping off, and in the manga when Osaka says she wants to see her life flash before her eyes - and Tomo responds with "Don't do it, Osaka".
Osaka was bullied in Osaka city, in the school there. She says in the manga, looking sad, that they called her names there, like "moron". And she never says she misses her old school. It's never revealed why her family moved away from Osaka city to Tokyo, but we can surmise it may have something to do with Osaka being bullied -- maybe life in general was bad for her there, and her parents wanted their daughter to grow up in a friendlier and better environment. That a reason for the move to Tokyo isn't provided -- while at the same time Osaka being a transfer student and having moved is made a big deal out of -- that in of itself can point to Osaka being bullied being the reason, as a short comment like "My dad recently got a job in Tokyo so we had to move" could have been thrown in somewhere but wasn't.
In her new school her classmates are friendly with her and supportive, but at the same time they nevertheless point out to Osaka that she's slow and so on, even going so far as saying there's something wrong with her brain. How does she really feel about that, getting told that sort of things? For the most part we don't see Azumanga from Osaka's perspective or hear her thoughts etc -- most of the scenes, we see only as outside observers. But it's logical to assume she doesn't like being told she's slow or dumb. And it's also logical to assume she feels inferior and different (in a bad way). Because objectively she is in many ways inferior, both physically and mentally.
Regarding physical inferiority, for example: In the manga Sakaki tells Osaka her large breasts aren't really that impressive, that they're just big because overall her body is bigger. She tries to make Osaka feel better about herself through that argument - Osaka responds saying "Oh so if we were the same height, we'd have the same breast size?" and Sakaki nods her head, then Osaka looks away, looking sad, and says "Yeah right...". And later, in the anime (maybe in the manga too, can't remember), when they're in swimsuits and she talks to Kimura and he ignores her, she then moves her hand over to her breasts and does a "grabbing" motion with her hand, showing that she realizes Kimura ignored her because she wasn't worth talking to because her breasts are too small. And she performs very poorly in sports and gymnastics. The point is Osaka is obviously aware of her inferiority and undoubtably feels inferior in that regard, as well as mentally/intellectually.
Aside from that, you should notice that a lot of the "jokes" Osaka makes or that are associated with Osaka involve death or things which would kill her. Some of those scenes you can read deeper into or extrapolate things from. But that's not what I mean here. Primarily what I mean is that you should notice that thematically, one of the themes around Osaka is death, dying and dead things - that it's a recurring theme. For example she looks at the skeleton of the Yamapikarya cat and sounds downright happy when she says the cat will one day look like that and thinks it looks "nice and bony" (regarding that, why she said "nice and bony", here we can think a bit deeper -- all things considered, did Osaka have an eating disorder? did she starve herself at some point? is that why she's physically the way she is? did malnutrition stunt the growth of her breasts and make her weak? she actually says, to Kagura, that she eats very little). And when she's in the pool she talks about how in order to float "all i have to do is become a dead body". And then there's the "we will fill the place with animals... and they'll all be dead" scene. And the scene where there's a ghost in one of Sakaki's photos and Osaka says she knows the ghost. And then Osaka's horror story in which she kills her classmates. And then there's her dream where she kills Chiyo and takes her pigtails and flies away crying. And there's the pool scene where Chiyo looks like she's drowned and Osaka looks happy (yes that's because to Osaka "Chiyo learned to float", but the point is: it visually looks like death, and Sakaki is terrified but Osaka looks happy, as if she's happy Chiyo died (and no I'm not saying she thought Chiyo was dead or that she was happy that Chiyo looked dead, I'm merely saying that's how the scene is presented visually). And then of course the scene with her thinking of jumping off the roof, and wanting to see her life flash before her eyes -- things that would in effect kill her. Those are the ones I can think of right now. Then you have the odd scene in the anime with her standing alone on the sidewalk with Tomo and Yomi behind her and Osaka turns around and says, looking serious and strangely sad, "I will never forget you". That scene is quickly transformed into a funny lighthearted one (hearing Tomo speak made Osaka happier), but, regardless, she was standing still on the sidewalk alone in some deep thoughts which lead to her saying that; so what was really going on in her mind? The point here is that the first part of that scene is odd and seemingly unfitting for Osaka -- if one views Osaka as being only what she appears to be on the surface, that is - but if the viewer recognizes Osaka as being troubled and having dark thoughts, then it's not strange at all.
And while just a small detail, it's interesting and relevant when everything else is taken into consideration: Osaka's eyes are different from everyone else's. The shine in everyone's eyes is located in the upper region of the eyes, so the eyes are "pointing" upwards, they are "uplifting" visually. Except on Osaka. On Osaka the shine is located in the lower region of her eyes, so they "point" downwards.

And I dont think people are twisting this show into "an edgy fanfic", this is looking for meaning. Even overtly comedic shows like Spongebob have a bit of meaning in them, sometimes quite deep if you're willing to look into it beyond surface level.

Out of all relationships in the show, chiyo and Osaka is by far my favorite. It's painfully sweet. We can tell how Chiyo cares for a big deal for all of her friends (even tomo), but the care she has for Osaka is a bit different. It's like she should be the younger sister to her and she behaves like she's the older 😂

this scene reminds me of a recent field trip i made. at the end of the day, we were supposed to make a picnic at a really beautiful place that was like a giant hill. me and my friends were looking the landscape at the top, really close to the end of it (of course there was some kind of railings, but it was pretty easy to jump off of it if wanted to) and i said something like "it's so beautiful and high, i wonder if my bones would break if i jumped off right now and what would be the sensation; i guess it would be an exasperating yet liberating experience". to the rest of the day they started treating me a lot more sweetly.
