Yes you can. There's a device you can add to your helium regulator called a 60/40 which automatically fills with a combination. There is a time when you cannot mix & that's when you are stuffing balloons with smaller balloons - they both must be all air or all helium.

Cotton, nylon or you can just use your hand.

Yes it does but not as well as foil. In the video I mixed both.
