Can i Do a flow cell / pore check directly after adding the storage Buffer? Because i want to see how many pores are left before putting the flow cell in the fridge.

So, if by any chance your flow cell check returns zero pores, you can solve it by starting a sequencing and letting it run for some minutes. So the software will return the right number of the pores in the flow cell.
When you are re-using the same flow cell for another run, are you suppose to wash the flow cell again before priming the flow cell? Due to storing with storage buffer?

If you are going to reuse the flow cell in another sequencing run immediately after its use, you should wash the flow cell with DIL + WMX, but it is not necessary to add the storage buffer (S). Then, proceed with the priming of the flow cell and load the library. But if you will store the flow cell, even if for some hours, I recommend adding the storage buffer. Hi when I use storage buffer it normally takes a couple days to get an accurate pore check
