    If you did not know this,you can try. Use your hand and with thumb and forefinger squeze them and try to make miniest hole in center of forefinger,go to light and look close,you can see bacterias in your eye. And if you look at screen it would zoom in a little bit.
    I dont know how i discover this,but its really amazing

    In our clg they thought like 9 drops of acetocarmine stain and one drop of hcl and heat until few minutes and put the heated stain one drop on sample using blotting paper press it and observe it but i am not understanding which method to use sir..

    Can I use methylene blue instead? I don't have acetocarmine that's available.

      Sir. Acetocarmine is use to stain chromosomes only, it is useful for visualization and position of chromosomes, methylene blue will stain all cellular material and chromosomes will not differentiated.

    I try to do this for thesis experiment but i encoutered many problems to visualize chromosome clearly at 100* even i tried it with two different stains acetocarmine and hematoxylain !? I don't know where is the problem???

    We not used dil HCL but we used conc hcl sir

    Why acetic acid is not used?

      Mitosis is continuous process, Acetic acid is used to fix root tip cells temporarily.
      While experiment dilute HCl is usefull to dissolve cell wall and cell can be separated.

      Treatment of HCl is very sensitive, it should be properly warmed. Not less, not more.
