To put this piece into context:
This the final boss theme of the newest raid in Destiny, where we infiltrate a sunken Pyramid ship, essentially one of the fleet of our main enemy. This boss is the First Disciple of essentially our biggest antagonist yet and a leader of the Darkness. This boss single-handedly genocided his entire race, subjugated several others, and is(or, was) arguably one of the most powerful beings in the canon as of right now.
Typically, Destiny tracks have a part that plays once the boss enters its "damage phase", in this case beginning at 3:47, where one of the main motifs of the game EXPLODES into the song. As you say, the track changes at this point as this is the point when the Guardians(us!) start to gain the upper hand. We've gotten past the Disciple's tricks and mechanics, and now it's time to counterattack. This is genuinely one of the best raids to come out and it was nice to see your breakdown.

Also, it is important to note that the "Darkness theme" we hear in the first few seconds of the song is more triumphant, mean, and intimidating, instead of the mysterious evil theme we have heard in the past, when the Darkness was still unknown besides the pyramids. It represents that the Darkness is HERE. NOW. Basically, the Darkness saying, "I hope you're ready Guardian, because the time has come." I love Bungie's musical storytelling so damn much.

You can also hear how at that section the strings actually take the spotlight with the guardians/lights motif. But at 4:46 brass section recovers, aka how Rhulk regains strenght and we go back to the symbols phase. This time they add the choirs.
Also important to note one thing he says, “The drums keep it constant and together” and it’s basically the dichotomy we’ve seen in the game “The line between light and dark is so very thin” or “light and dark are just faces of the same coin” that’s why I believe in some point brass is the counter point and other moments strings are the counter point.
And let’s not forget that damn intro, massive intro, is the same darkness theme/motif we’ve been hearing here and there, first was really subtle and here it finally is present without distortion!!

I love how bungie's music team and composers blend and manipulate their music so well. The boss music doesn't play from start to finish if say you where just listening to it on UA-cam or a download. The parts of the song typically play depending on the action currently happening in the boss fight. Examples include what phase of the boss you're at, are you in the dps damage phase of the boss as well as you just beat said boss etc. Hope that makes sense. Dynamically playing parts of the song is the best way to explain it.

So synthetics (to my mind) usually come with a bit more meaning for the Destiny franchise, same with strings. Synthetics often come in representing the Darkness and its followers, while strings refer to the Light and its followers. Music actually plays a really important part in Destiny’s world. like Savathun’s song was used to cause a political uprising in the main city, and allow alien robot zombies in. There’s also the death song where anyone who hears it dies, (you actually see it’s effects in a lore book and it is terrifying) that song was also once a song of life corrupted at some point.

The intro that you highlight is the motif of the Darkness and has shown up quite a bit as it's taken centre stage, while the heroic sections of the piece are a motif from 'Guardian,' a track off the soundtrack of the original Destiny that's been with us for more than seven years. The Guardian motif even appears in the tracks that you've previously covered on this channel.

The namesake is for Rhulk, The First Disciple. He committed genocide on his race because he felt like it, then exterminated several other races also because he felt like it. Among the several literal gods, mechanical abominations, and reality-bending dragons we've clapped, the First Disciple to the Witness is by far the most powerful and menacing enemy Guardians have faced.

If you hear the track you will also distinguish two different rhythm. The first rhythm is like a dance cause you have to do a mechanic to not get killed and is a very schematic mechanic in which you have to keep a constant rhythm in your action. Later on the boss accepts you to a direct melee fight where you have to break his armor and after that start the real beatdown between the boss and the team. In the second phase the mood of the boss change from "let's see how good you are" to "how the f*ck you dare challenge me you foolish humans". It's a very well done soundtrack for an amazing boss fight

All the motifs give me chills.
I know you’ve got it pretty much correctly. The mean brass section it’s the trademark of the “Darkness” or the main antagonist of the game. During several years that motif that plays at the very start was hinted in several parts of the game, muted, deformed but each time being more and more defined till this theme came out, then, the trumpets/violins play for the guardians/light (us players, heroes and our power) and the choirs are for the traveler itself (our good side god).
You can get the moment when string section dominates, that’s when the player starts to win, then brass regains energy (as boss fight goes back to pre-dps phase)
You at the end can hear the strings and brass talking to themselves, basically in destiny terms, guardians win by mastering both sides dark and light. And the synthetic part, that synthetic part is The Witness(the boss of this boss, the real bad guy) trade mark, d2 players will remember that ringing everywhere.
The layers being stripped it’s for the gameplay purpose when players are between phases to “give a break”

You’d also hear the (Ambient) and (Tension) layers, these are used to represent your phase in the encounter as things are changing. Drown in the Deep has great (Ambient) and (Tension) pieces, and every part of the action for the 3 phases is very satisfying.
In this encounter there are three phases, and the in-game version sounds much different, starting to add instrumentation layers over time before resetting for the next phase. At the start of the succeeding phases (of which there can be a maximum of 3 for this boss) the music will 'grow' even more between, so it's not a full reset. At the low end of his health bar, he enters 'final stand' in which the music becomes "the most intense".

I’ve listen to this particular track many times, and this video as well. What’s really awesome about this piece is that it describes the flow of the conflict between the two participants from start to finish.
The brass and synthetics represent the Darkness (mainly Rhulk, the titular First Disciple of the Witness) and the strings and other instruments are the Light (us players, the Guardians).
Both of these motifs are trading melodic jabs at each other all while the drums maintain the pace of the fight throughout!
Funny that you mention how spacious the song is, because this occurs inside one on the Pyramid ships associated with the Witness; a cavernous, almost hollow interior with very few structures to navigate throughout.

the coolest part of Sepiks Perfected is it was a fan cover that bungie asked if they could use in the RoI version of devils lair :)

Little tid bit of info about the climax of the song with the really triumphant brass section, but those are the same notes with only slight variations as the Vanilla D1 main menu theme called “Guardians” (OST 7).

Day 67 of requesting House of Sacred Remains from Castlevania: Lament of Innocence. Beautifully atmospheric track and one of my fave's from video games in general. It'd be awesome to hear what you think of it.
It's the theme of a level set starting from a cathedral and ending in the crypts through lots of library areas, it's not a particularly significant or infamous level at all but I think the track really reflects the hallowed grounds housing sacred relics beautifully.
The 4 minute upload from the Castlevania Sound Team is the one to listen to.
