    Dr. James Kirkland, Mayo Clinic: Clinical Trials & Senolytics
    James L. Kirkland, M.D., Ph.D., is Director of the Robert and Arlene Kogod Center on Aging at Mayo Clinic and Noaber Foundation Professor of Aging Research. Dr. Kirkland’s research is on the contribution of fundamental aging processes, particularly cellular senescence, to age-related diseases and development of agents and strategies for targeting fundamental aging mechanisms to treat age-related chronic diseases and disabilities.
    Additional research areas include molecular and physiological mechanisms of age-related adipose tissue and metabolic dysfunction, frailty, and loss of resilience to acute diseases in old age. Dr. Kirkland’s laboratory published the first article about agents that clear senescent cells - senolytic drugs. Dr. Kirkland demonstrated that senolytic agents enhance healthspan and delay, prevent, or alleviate multipleage-related disorders and chronic diseases in mouse models. He published the first clinical trials of senolytic drugs and is currently conducting multiple clinical trials of senolytics. He has more than 200 publications and holds over 20 patents. Dr. Kirkland is Principal Investigator of the Translational Geroscience Network (R33 AG061456), which brings together 8 academic institutions to translate healthspan interventions, including senolytics and other drugs that target fundamental aging processes, from bench to bedside. He is a scientific advisory board member for several companies and academic organizations. He is President-Elect of the American Federation for Aging Research, a past member of the National Advisory Council on Aging of the National Institutes of Health, past chair of the Biological Sciences Section of the Gerontological Society of America, and a member of the Clinical Trials Advisory Panel of the National Institute on Aging. He is a board-certified specialist in internal medicine, geriatrics, and endocrinology and metabolism. Dr. Kirkland is the 2020 recipient of the Irving S. Wright Award of Distinction from the American Federation for Aging Research.es.     It's understandable to not have all of the answers in such an incredibly complex process. It's however very exciting for those of us suffering from chronic illness. Thank you for sharing.

    I've been taking quercetin and fisetin off and on for a long time now. No side effects and one extensive blood assay showed low inflammatory markers. Waiting for Mayo to catch up, followed by FDA 25 years later.

    Last time i listened it 12 years ago
    I want to live forever young, which are the most gorgeous days of my life (full of dapamine and oxytocin)
    I can't express my feelings in words, i don't even know english , but hey i know you are understanding me, yes, I'm talking to you from bottom of my heart, I'm broken, i want those days back, i know every life lesson knowledge, but.... I can't

    There was a paper in 2019 by Grzegorz Grynkiewicz and Oleg M. Demchuk, titled New Perspectives for Fisetin, who found that Vitamin C enhances the anti cancer effects of fisetin. I wonder if this would apply generally to its senolytic effects, and whether Dr. Kirkland would consider adding Vitamin C to fisetin in one of his clinical trials?

    Thanks. Very informative. It was interesting to note that recent work out of the Conboy lab showed that (in mice) large dilution of blood plasma (50% replaced with saline) reduced SCs in the brain by a huge amount. What is not known is how long the effect lasts, and what is the minimal effective dose as there was no apparent dose response curve developed for plasma dilution. Blood bank plasma donation will remove 20% of plasma for example. Need SCs testing for fasting regimes, exercise, diets, as well as natural substances like fisetin and quercetin. A lot of unknowns to sort out.

      Peter, I am assuming the plasma in the elderly contains much of waste / harmful substances from the liquid portion of blood, so Therapeutic Plasma Exchange removing senescent cells, reverses Alzheimer/ Dementia and reduces amyloid plaques systemically. The plasma dilution Irina Conboy, spoke of for Therapeutic Plasma Exchange, has Dobri Kiprov using the Spectra Optia Blood Cell Separator lasting 2-3 hours.So calculate the time for a "Blood bank plasma donation", and that will show the ratio of removal.

      @@garvintimmann Thanks. I think one TPE session removes about 50 to 60 percent of plasma. I started with plasma/platelet donations recently and learned that blood bank tracks amount of plasma loss each session and limits it to 13 liters a year over all donation type. So I will be bale to keep a good record. The most I can lose in one session is about 20% of plasma volume. About 10% for single platelet donation (can be done every 2 weeks) and in between for combination (done every 4 weeks). In any case, the maximum monthly plasma loss due to various kinds of donations is about 20 to 25%.

      @@fondrees maybe check albumin which is normal part of cheap routine bloid work. Optimum is above 4.5. Don't want it low. If so you could go to once a month donation.

    Just ordered Fisetin from Do Not Age, bought some Quercetin from Whole Foods. I’m 35, fit, no health conditions, but I’m a health/longevity junkie. I’m going to go on a 5-day quercetin/fisetin pulse cycle once a month.

      Please let us know the outcome. One thought is we do not know if Quercetin and Fisetin work together or inhibit each other. Your input may give a clue.

      I just tried a hit and run with two fisetin doses. No side effects and I will probably never know if it worked. Except by following the clinical trials.

      @@y.g.1313 Alive and thriving. I'm feeling much better lately, but I can't confidently say it's because of the fisetin as I've done a 5 day fast since I made this post, I've also cut my saturated fat intake. But I can say with reasonable certainty that the fisetin didn't cause me any harm.

    Is there a timeframe after senescent cells appear that you have to introduce rapa/etc to clear them out? Or can you do it at any point before cancer? For example, if you got a CT scan 10 years ago, can taking rapa/sirolimus now clean up those old issues? Or need to pulse like immediately after?

    When he says not to use "these drugs" yet is he referring to things that would require a prescription anyway or Quercetin and Fisetin which are supplements that can be found in nutrition stores?

      These "supplements" are being used as drugs to attempt to treat age related health conditions in high doses. Hopefully the FDA isn't going to try to control these substances.

    Dr Kirkland. Is saying that he's going to try to take fisetin a natural bioflavonoids sold in stores off market as a drug. My mother been taking. QUERCETAN and vit D. For 25 years where has Dr Kirkland been ??

      There is not much actually, just the usual 'we are in phase 1 or phase 2 trials'. At 16:00, there is a list of prospective 'senolytics' tested and it's Fisetin and Dasatinib + Quercetin. Went to ClinicalTrials dot gov, and they are still under the 'recruiting' phase, so I think we wait a few more (like 5 or 10) years

      @@peterng25 fasting/FMD is available today as senolytic therapy. Check out Valter Longo if interrested.
      Also quercetin and fisetin is very available, “biohackers” take them.

      @@iblisthemage Please give me a hand, cause I am confused ☺️If I am 80kg, and as per Mayo Clinic protocol I have to take Fisetin 20 mg per kg- it’s -1.600mg. So if one pill is 100 mg the I will have to take 16 capsules a day for 5 days a month🙏?

      this is good Jambu ! look at it this way: if you really fear something- you make more effort to avoid it? yes.. so: live 10-20 yrs longer, that's not bad for starters.. then- solution will be available (death problem will be solved)
