    WILD! I heard about CO2 Enrichment in Greenhouses, but to see it at a Feild/Forest Scale is amazing! Granted it’s for scientific data gathering, not for increased growth, but it is interesting to see the concept scaled that way. This is definitely a rabbit hole I’ll dig into!

    So, more CO2 is better for most plant life. That's better for herbivores. Clover etc fix nitrogen. Farmers add CO2 to greenhouse spaces; they're not wrong. Missing statistics -> no evidence -> what confirmation can anyone else draw? Poor scientific disciplines, unfortunately. Rising 'average' temparatures come before rising CO2. Tiny fraction of 'warming' is from CO2 and tiny fraction of that is man made. Panic over.

      I'm not sure exactly, but it is some combination of plastic film and sealing tape. These samples on the bench top were for demonstration purposes, but depending on what you are studying, you might need to ensure an airtight seal or leave the bottom of the columns open (e.g. if you are in the field).

    Congrats fo the video and thank you so much! A couple of questions arise for me: how is the amount of C analyzed in each fraction? Can be used an elemental analyser for this purpose? If so, how to separate organic carbon from mineral carbon? Thanks a lot

      You can separate mineral and organic carbon with the use of 2 analyses, separately. This this may lead to the need for a larger sample mass (at least 2 grams, better 5 g). So, you measure carbonates with calcimeter and determine mineral carbon (Cmin). You subtract Cmin from Ctotal, which you measure on CN analyzator by combustion at 900 °C. This is one way to get Corg.

    Stereo microscope. :) I have seen two videos where a stereo microscope is used, and one with a compound microscope. Soil people, what is you preference?

      Binocular should result in less eye strain over time. I'm only looking for binocular or stereo myself, but would use a single eyepiece unit if I had access to one.

      They sink in pure water, she said they float when the density matches. So more sugar is probably better. Fill it to capacity until it doesn't dissolve anymore.

    Thank you for the video tutorial. I have some questions. 1) How long(approximately) will it take to analyze 66 soil samples by your methodology? 2) Are there any possibilities to decrease used water amount in fractionation?

      Hello, it has been some time since I have done the procedure, but if I recall correctly, the maximum number of samples we would process in one "batch" was about 16. Any more than that, and it is difficult to finish each step in one day. We generally took three days for the whole procedure, so it would take about 9 days in total for you. With practice, you might be able to decrease water consumption during the rinsing of the LF and the sieving, but you need to make sure you are being thorough. Also, if you do change the amount of water that ends up going to the centrifuge (or use different sized bottles), you will need to calculate the speed and time of centrifugation to separate the correct size particles using Stokes' Law (en.wikipedia.org/wiki/Stokes%27s_law).
