How to grain to grain transfer| Fastest colonization method
Check out our video on how to make your own Master's Mix 5lb block: ua-cam.com/video/y2I9bZppPQk/v-deo.htmlsi=ZyC-NHJ1cNRHcBeZ
Materials Needed:
✅ Fully colonized bag or jar of grain spawn
✅ Sterilized bags/jars of grain (receiving containers)
✅ Alcohol spray (70% isopropyl)
✅ Sterile gloves 🧤 / face mask 😷
✅ Laminar flow hood or still air box (SAB) 🌀
✅ Measuring utensils for transferring grains (or eyeball 👀 if comfortable)
✅ Impulse Sealer 🔒 / Stapler 📎
Procedure:
1. Preparation: 🧼
Put on your mask 😷 and gloves 🧤.
Clean your workspace thoroughly 🧽.
Use a laminar flow hood 🌀 or SAB to maintain sterility.
Sanitize hands, tools, and the exterior of your colonized spawn bags/jars with 70% isopropyl alcohol.
2. Open Spawn Bag/Jar: ✂️
Carefully open the colonized bag (using scissors ✂️) or jar (untwist the lid) inside the sterile area.
Avoid shaking it too vigorously to prevent contamination 🚫.
3. Open Receiving Bags/Jars:
Carefully open one sterilized grain bag 🛍️ or jar at a time 🕒.
4. Transfer Grains:🌾
Use a sterile cup/tool 🥄 to transfer 1-1 ½ cups of colonized grain into the receiving bag/jar.
Distribute colonized grain evenly for quick and uniform colonization.
If comfortable, eyeball 👀 the measurement.
5. Seal and Shake: 🤝
Reseal the receiving container immediately with an impulse sealer 🔒 or stapler 📎.
Gently shake or roll the jar/bag to distribute the colonized grain evenly 🤲.
6. Repeat: 🔁
Repeat the process for additional jars/bags until complete.
7. Incubate: 🌡️
Place inoculated grain jars/bags in a clean, dark area 🌑 with temperatures between 70-75°F (21-24°C).
Allow 7-14 days for full colonization ⏳.
Tips for Success: 🌟
Minimize airflow in your workspace if using a SAB 🌬️.
Don’t overfill receiving jars; leave space for shaking.
Always sterilize tools between transfers to avoid cross-contamination 🚫🦠.
Discard jars with visible contamination ❌.

It's a mycology thing, we cannot help if people have an issue with needles, Liquid Culture (needle juice)! We have been a business for a year and you're the first person to mention such thing.

Are your jars sealed up tight or do you have air holes in top?

Each jar has at least 1 quarter inch hole covered with either micropore tape or filter patch in the lid for gas exchange. 😊

Three hours in a pressure canner is a good way to run dry and warp your canner. Ask me how I know! 😅

If you put the right amount of water in there you will never run dry lol, its crucial you put enough water when running your PC for 3 hours but this is proper sterilization time for oats :)

@ I always add 5-6 quarts of water, but my canner is old and allows more steam to escape than it should. And it’s much worse now that it’s a little warped! 🤪 Perhaps better canners can go this long, but I will never go that long again with a Presto. I’ve been PCing my groats for 90 minutes, and that has been successful for me! I think having them properly hydrated beforehand is the key to ensuring proper sterilization, but to truly sterilize them, you need an autoclave.
