Thank you for showing us the slow shaking! so this is for sample and Ab binding ( slow shaking for 30 min & incubation overnight)? I wonder if 400rpm does not make any sense to you... I am new to ELISA but i did some WB. Abcam recommends incubation of sample and Ab at room temp for 1 hr with shaking at ***400rpm*** i followed the protocol and the highest abs signal only reaches 1.78, which is 1/2 lower than the reference datum Abcam provided. I think it's the aggressive shaking reduce the binding causing low signals. I am going to contact the customer service too. Thank you in advance!
