Frederick National Laboratory for Cancer Research

The Frederick National Laboratory for Cancer Research, currently operated by Leidos Biomedical Research, is a U.S. national laboratory focused on discovery and innovation in biomedical science and rapid response to emerging infectious diseases. Sponsored by the National Cancer Institute, the laboratory collaborates with public and private partners to solve urgent and intractable problems in biomedical science.
Biotech Connector: Structural Biology as a Tool
Speakers at the August 22, 2024 Biotech Connector event shared how structural biology has enabled them to better understand molecular mechanisms and explore therapeutic innovations.
Ruth Nussinov, Ph.D., senior investigator and head of FNL’s Computational Structural Biology Section, discussed using structural biology to better understand the pathways where such mutations arise, enabling investigators to select better combinations of cancer therapies to use against mutations.
Carter Mitchell, Ph.D., chief scientific officer of Kemp Proteins, explained how a fascination with structure sparked his interest in the biological sciences, and how structural biology remains key to his current work elucidating protein functions.
Manu Kohli, Ph.D., principal scientific advisor at Charles River Laboratories, presented on how mass spectrometry techniques can support drug development.

Can you explain what happened?
The HNH and RuvC domains are the nuclease domains responsible for cleavage. A lot of Lys, His, Asp, and Glu residues are responsible for binding Mg2+ and facilitating phopshodiester hydrolysis

Cas9 is a nuclease(it can cut DNA or RNA strand)enzyme which is combined with a tracerRNA(which helps to bind to your target sequence ). This cas9 can cut because it has two domain( area) called 1.HNH and 2. RuvC-like domain.When cas9 binds to a target DNA sequence, its HNH part binds to the strand of the DNA that is complimentary to the tracerRNA, while RuvC-like domain binds to the non-complimentary strand. These two domains will cut both the strands of your target DNA.
