Excision BioTherapeutics could make adjustments, utilizing CCR5-delta-32 (CCR5Δ32) as a vector for EBT-101. Excision BioTherapeutics' EBT-101 was very close to establishing a possible cure from HIV, with sucessfully passing Phase 1 human clinical trials and the FDA approving and granting Excision BioTherapeutics' EBT-101 an FDA Fast Track Designation, but unfortunately EBT-101 was proved to be "ineffective against HIV", during human clinical trials, while stem-cell transplants via CCR5-delta-32 gene mutation has cured seven people in certain situations. What is the possibility of utilizing CRISPR-Cas9 to deliver CCR5-delta 32 (CCR5Δ32) with guide RNAs into human cells? EBT-101, still has potential as an HIV therapeutic cure, with EBT-101 having an approved FDA FastTrack Designation. While EBT-101 with it's current make up, being well tolerated in patients, in intial human clinical trials, unfortunately EBT-101 failed to eliminate HIV, during clinical further human clinical trials. For clarification EBT-101 is CRISPR-based, with the utilization of 2 Guide RNAs to attack HIV. So far [1] the late Timothy Ray Brown, the "Berlin Patient" (who passed away in 2020 from a returning leukemia not caused by AIDS), [2] Adam Castillejo, the "London Patient", [3] an unnamed mixed-race woman from New York, [4] an unnamed 'Geneva Patient" Swiss man, [5] Marc Franke "Dusseldorf Patient" 56 year old German man, [6] Paul Edmonds, the 68 year old "City of Hope Patient" and the oldest people that had completed the CCR5Δ32 stem cell transplant, and most recently [7] a 60-year old man in Germany, was announced to be free of HIV after receiving the CCR5Δ32 stem-cell transplant, which the 60-year old man has been HIV-free for close to six years after receiving a stem-cell transplant from a donor with only one copy of the CCR5Δ32 mutated gene. This latest case of a person receiving a sterilizing HIV cure, means their cells do express CCR5, but at lower levels than usual. As presented by Ravindra Gupa, a microbiologist at the University of Cambridge, UK, and stated by Infectious diseases physician and Melbourne l, Australia Head of The Peter Doherty Institute of Infection and Immunity, Sharon Lewis, this recent case sends a clear message that finding a cure for HIV is "not all about CCR5". This recent findings widen the donor pool for stem-cell transplants, which is currently a risky procedure offered to people with leukemia but unlikely to rolled out for most individuals with HIV. Roughly, 1% of people of Europe descent carry both copies of the CCR5Δ32 gene mutation, but some 10% of people with such ancestry have one mutated copy. Also, this latest case "broadens the horizon of what might be possible' for treating HIV, says physician scientists Sara Weibel, who studies HIV at the University of California, San Diego. This only strengthens the possibility of a current possible HIV therepies currently in eight-stage clinical trials, in which the CCR5 receptor is sliced out of a person's own cells using CRISPR-Cas9 and other gene editing techniques, says Sharon Lewin. Even, if these therepies don't get to every single cell, they could still have an impact Excision BioTherapeutics was reasonably close to being FDA approved. In July 2023, the FDA granted approval for Excision BioTherapeutics' CRISPR-based EBT-101 to receive a FastTrack Designation, and the results of the EBT-101 Phase 1/2 human clinical trials was a success. There were no reactions related to participants with HIV taking the EBT-101 infusion and no complement-medicated toxicity. EBT-101 was detected in their blood four weeks after infusion and all three participants, and Excision reported no evidence of the gene vector in tissue associated with male reproductive function. EBT-101was developed and researched by Excision BioTherapeutics, at Dr. Kamel Khalili's lab at Temple University and Jennifer Doudna’s lab at UC Berkeley, is a CRISPR-Cas9 and two guide RNAs (gRNA), with the purpose of delivering the one-time treatment into cells via an adeno-associated virus (AAV), that targets three (3) sites within the HIV genome, and cuts out large portions of the HIV genome and minimizing potential viral escape. EBT-101 currently in Phase 2 (II) human clinical trials. As previously stated, the clinical trials for EBT-101 encountered an unfortunate setback, as reported from Excision BioTherapeutics in March 2024, that while EBT-101 was safe and well tolerated from participants, EBT-101 did not prevent HIV viral rebound and failed to maintain viral suppression, when used alone at the initial dose tested, in three patients who stopped antiretroviral treatment in an early study. EBT-101 may have only delayed. In theory, this approach of EBT-101 accompanied with CCR5Δ32 as it's vector would have EBT-101 employing CRISPR-Cas9, two guide RNAs, along with CCR5Δ32, which would target three sites within the HIV genome, thereby excising large portions of the HIV genome and minimizing potential viral escape, while preventing HIV from replicating, by blocking the receptor on CD4+ immune cells that HIV uses to enter and attack the immune system.

can we have an update about AGT they are asking for phase 3 . when did the second happens and what are it's results

i've seen this video earlier and i had hope . lately i've been seeing a back up from doctors saying that no we will defeat hiv by stoping the spread but the cure is later on . i really hope the treatment will be oit by 2030 . i will be so happy . yes i lost my 20 years because of hiv and depression. cartilage damage as side effect .but i'll make up for it all after ending the stuffer from hiv treatment. i'll do lot of stuff m god pleaaase help them and ease the research for them guide them to the solution easily
