      You have to allow room for shaking/breaking the grain. If you overfill it becomes very difficult to shake. This is how we refer to spawn volume. The actual volume isn't an entire q, we use the jar size as a reference.

    Would you please test 2 more jars with 0.5 inch of water; one with tightened [2 peice ring] lid; one with a "finger tight" lid? I'm being told on Reddit that you have to vent the trapped air **in the jar** to achieve sterilization. I believe that the venting is to allow a vacuum to be formed after cool-down. Some mycologists find undesirable because it sucks in "dirty air" (we'll ignore the flow hood) when opened. I believe that a tightened lid will still sterilze, but result in no/little vacuum after cool-down. The lids and jars should be okay (unlike in your dry test) because the pressure inside the jars will increase because the water will turn to steam.

      Corn is very clean but is too large with too few inoculation points for my taste. Oats can be troublesome, it depends on the source. I don't recommend either.

      Huh? I have not had a problem with oats. Trying to see of going to corn was worth it. Doesn't sound like it though. I rarely have contamination issues. Can I ask what your grain preference is? Seems like my cultures actually do better when I switch things up once in a while. Not wbs or rye thought. Rye is kind of a pain to get right and is ridiculously expensive here. Everyone keeps talking about this drippy tech which seems pretty dumb but you kinda confirmed my suspicions. Not worth it really.

      Because coir/vermiculite that has been expanded via hot water has no contaminant organisms. Competitor organisms will piggyback on contaminated spawn, infecting the substrate. This is why clean spawn is so important. This is not the case however with manure substrates that can mold when improperly processed.

    I tried repeating what you did. Right now my substrate is 90% colonized and there is no colonization on my casing layer. Visually the layers are very distinct. I'm concerned that I may have packed my casing layer too compact. Do you think that's a possibility and has that happened to you?

      Essentially you need enough to not run out while not submersing the jars. Steam under pressure reaches higher temperatures than the boiling point of water which is where the sterilization comes from.

    Very informative, thank you. I've watched a lot of videos on mycology on youtube recently, and yours is the only one to cover the reasons not to use to a glove box. Lots of people promote SABs, but don't mention glove boxes. Could be very easy for a noob like myself to see a glove box, and think it's a better/upgraded version of an SAB.

    Great videos man, clear and concise. My only suggestion for a video like this though is that it seems a bit incomplete. How are you sterilizing the scalpel? Do you sterilize between each transfer? Are you wearing gloves? I'd love to see your full process

      Few people engaged in this hobby are running IR units, they're a bit different than flame sterilization; but yes, flame sterilize until red hot, and cool in the receiving dish. The intention of this video was to show how and where to place dishes in relation to receiving jars, it was not intended to be comprehensive.

    Purging the pressure cooker? I’ve never done it and will never do it and have always had stellar results.. people need to drop all these old-school methods. Growing mushrooms is quite easy. It’s just very tedious no need to add a bunch of extra mumbo-jumbo to it. Not saying to cut corners, but some corners shouldn’t be there.

      It's not "old school", this is how autoclaves function. Dalton's law covers this phenomenon: Quote: According to Dalton's law of partial pressures, the total pressure by a mixture of gases is equal to the sum of the partial pressures of each of the constituent gases. The partial pressure is defined as the pressure each gas would exert if it alone occupied the volume of the mixture at the same temperature. If you don't vent/purge the pressure cooker or autoclave you won't reach the appropriate internal temperature.

      It’s a personal preference I’d recommend using brown rice since it’s a lot cleaner than feed grain

    Hi, thanks for this valuable info! And just starting and I'm going to remove the gloves from the holes of my SAB (it's just an inverted tub). I'll Also add something to put my materials a couple cm above the floor of the SAB. I wanted to ask you, if I Get more serious in the future, do you think making a proper SAB like this would minimize contamination reliably (providing good sterile technique) or is a laminar flow clearly better?       Both have advantages and disadvantages and can have different applications. Working with contaminated wild cultures or spore printing is best done in a still-air environment. Still air is often employed in areas that don't have access to a cheap/reliable power source in developing countries. Laminar flow allows for freedom of movement, workflow, and a greater volume of work when the appropriate infrastructure is available. Having both available is the best-case scenario. Sterile technique is key no matter what you choose.

      @StipeCap thanks for this detailed answer. I'm just a beginner but I'm Also highly motivated. I'll see how I can manage to make myself a proper and big enough SAB for now. I'm also trying my Best with sterile technique, exposing grains and spores as little as possible to the air and not putting my hands above this materials. It's more challenging that it seems, isn't it? If I can, I would like to know your opinion about this: should I, provided I Get a big enough SAB, prepare the cakes inside of it or not? Because it seems overkill, but, at the same time, we're really careful while preparing the grains, but not as much with the bulk, and I happened to Get contams at the bulk colonizing stage.

      @martindamasseno5164 Still air and laminar flow are for the sterile culturing phase of cultivation. Producing pure culture on agar, the inoculation of media like spawn, liquid culture, grain to grain transfer, etc. Anything past that phase that doesn't involve nutrient-dense substrate or media can be handled in the open air.

      Start with a grow kit, read a ton of “shroomery” it’s a forum about growing in internet, start with little, and never do something without doublecheck in mushroom cultivation, I learned that the hard way

    Agree on simplicity of SAB and usage. On the part of adding fan and filter internally and gloves access, thats depends on use and environmental factors. Not to say it is good, just an added layer of getting better air quality control within the box. The inter filter and fan doesn’t need to be turn on during actual work, just need to pre-clean and turn off beforehand.

    Hey man, I appreciate all of your information and work. You do a really good job at explaining everything, thank you for that. I have a few questions that I hope you can answer. First, what is the dry weight of millet per bag before you hydrate them? Second, how many of your 5lbs sub bags are you able to inoculate with one of your grain bags? Third, would 14A .5 micron unicorn grow bags would work for this? Finally, I am using a gas stove. My PC is some brand that Canadian Tire carried a few years ago and is no longer available, i've had no issues with jars but do have some concerns that the hot metal could melt my bags. Would bringing the boil/heat up slower be the play here? This last one is a little vague, I may just have to mess around with it a few times but don't really want to dick around with a pressure cooker and stuff melting inside. Thanks again!

    Did you mix your grain with the coir or just leave the grain layer at the bottom & top it off with coir?

      Mixed into bottom layer, then topped.

    Another question. I just got my malt, but must have misread it as it is liquid malt extract. I am just going to experiment making plates, but do you have a recommendation on how much to use if I was doing the same as your video here?

      I've not used liquid so just play with the ratios to see what works best.
