Monitoring Painted Turtle Populations Post Wildfire
Following a 225 hectare Wildfire in a Jack Pine Sandstone Barren we monitored three wetland complexes for painted turtles using nylon hoop traps (Nylon Net Co.) in each wetland site every other week. All captured Painted Turtles were identified, uniquely marked and measured; All non-target species were identified, recorded, and released. We implemented filing on carapace to carve identification codes; Dial calipers were used to measure and record anterior and posterior lengths along plastron; Pre-cloacal tail length was used to determine sex; Right middle foreclaw lengths were used to determine age and gender. Water samples were collected and used to test pH, conductivity, and total suspended solids at each location.
This project was done under the supervision of Dr. Danielle Garneau through the SUNY Plattsburgh Conservation Biology course. DO NOT attempt this at home, without proper supervision, or proper permits.

4M KOH solution is not suitable for digestion . Because of same plastic types are dissolved in stron base or weight loss. So 10.% KOH solution is more suitable . And also some very important few steps were missed here.

We used KOH Digest and Wet-Peroxide Oxidation to isolate the microplastics, and then followed up with hand sorting/quantification with the use of a microscope.

@@aisselmontoya6392 The control is a sampling technique that involves selecting a sample in this case a beaker with no contamination like the beaker without the fish and only the chemicals to achieve a batch without bias. The sample is the beaker with the chemicals and fish. The chemicals will produce a reaction to the fish to produce a product. In this case digestion.

For this experiment we used hand sorting quantification and analysis methods with the microscope. FTIR and Nile Red staining are other methods used for quantification and identification!
It wasn't specifically a microplastic identification software but more of a general microscope imaging software. I took a look and it seemed like Leica microscope software

Very clearly presented. Can you tell me what liquid did you keep in the vial . How can we remove samples if it is small and transfer to the vial
The liquid in the vial is de-ionized water. Samples from the Petri dish can be transferred to the vial using forceps, using the microscope for visual aid. I hope this answers your question!

Hi Caleb! We used the Lecia EZ4W microscope which varies from 8x to 35x magnification. Our magnification varied based on particle size for optimal resolution. The imaging software was included with the microscope.
