    2:04 / 22:17 Alexander Comninos im surprised you'd make such a critical mistake with the Testosterone Range. Dr Tom Trvision who made it has never said that the range is healthy. Its only 1k people from Boston all exposed to industrial chemicals. Not to mention there are no population studies for dosing. Not to mention no body is doing body weight calculations like all other drugs are done. So sounds like you need to do combination dose studies from 100mg to 1G to 2G along side kisspeptin and see what works.

    Video summary [00:00:00][^1^][1] - [00:29:10][^2^][2]: Jason S. Carroll presents a comprehensive overview of estrogen receptor transcription in breast cancer, focusing on the role of the estrogen receptor (ER) and its interaction with cofactors and pioneer factors like FoxA1 and GATA3. He discusses the prevalence of hormone-dependent cancers, the effectiveness of drugs targeting ER-positive disease, and the challenge of endocrine resistance. Carroll highlights the importance of understanding where ER interacts with the genome and how it recruits cofactors to modulate gene expression, leading to cell cycle progression. He also explores the dynamic relationship between ER and progesterone receptor (PR), and how PR can influence ER's genomic binding sites, potentially offering new therapeutic opportunities. **Highlights**: + [00:00:00][^3^][3] **Introduction to hormone-dependent cancers** * Prevalence of breast cancer * Role of estrogen receptor in disease progression * Current drugs targeting ER-positive disease + [00:01:14][^4^][4] **Endocrine resistance and predictive assays** * Challenges in predicting treatment response * Progesterone receptor as a biomarker * New insights into ER and PR relationship + [00:04:01][^5^][5] **Estrogen receptor's interaction with the genome** * Importance of pioneer factors like FoxA1 * ER's dependency on pioneer factors for DNA binding * Potential therapeutic targets within the ER complex + [00:07:02][^6^][6] **Mechanisms contributing to endocrine resistance** * Various escape mechanisms in resistant models * The role of FoxA1 across different resistance contexts * The significance of ER mutations in metastatic cases + [00:10:02][^7^][7] **Dynamic relationship between ER and FoxA1** * ER and FoxA1's physical interaction and genomic binding changes * ER binding profile differences in tumors with good vs. poor outcomes * Progesterone's influence on ER binding sites + [00:23:57][^8^][8] **GATA3 mutations and their impact on ER function** * GATA3's challenging role and mutation prevalence in breast cancer * The protective nature of certain GATA3 mutations * Mutant GATA3's reprogramming of ER binding sites

    1. 400IU is too low to raise vitamin D level sufficiently. Why is the Society of Endocrinology pretending they don't know this? 2. Research on vitamin D as a treatment for Covid either used dosages that were too low or simply given too late. We don't research the protective effect of helmets by putting them on after a crash. 3. Virtually all medical research is directly is indirectly funded by pharmaceutical companies and their stakeholders. The incentive to properly investigate the effect of proper vitamin d supplementation on covid simply isn't there. This makes it all too easy to use "so far there isn't enough evidence to suggest..." as a cop out. Use common sense: we know hypovitaminosis D increases mortality and risk of serious illness and we know most people are deficient.

    Chronic subcutaneous long-term administration of 50 nmol kisspeptin-54/day for 13 days decreased testicular weight. Histological examination showed degeneration of the seminiferous tubules associated with a significant decrease in the circulating levels of the testes-derived hormone, inhibin B. Plasma free and total testosterone were also lower, although these changes did not reach statistical significance. Further studies examined the effects of shorter periods of continuous kisspeptin administration. Subcutaneous administration of 50 nmol kisspeptin-54 for 1 day increased plasma LH and testosterone. This effect was lost after 2 days of administration, suggesting a downregulation of the HPG axis response to kisspeptin following continuous administration. "Chronic subcutaneous administration of kisspeptin-54 causes testicular degeneration in adult male rats Emily L. Thompson *, Kevin G. Murphy *, Michael Patterson , Gavin A. Bewick ,"

      RIght. This is overstimulation of the HPG-AXIS. Interestingly, a similar degeneration can be found in men using human chorionic gonadotropin, or HCG. Kisspeptin-10 is not Kisspeptin-54. That distinction is important in the context of men using Kisspeptin to prevent exogenous testosterone induced testicular degeneration. The equivalent dose in micrograms, to the 50nmol of Kisspeptin-54, would be 65MCG. So - if one were to dose Kisspeptin-54 (which you cannot get for less than $700.00, and is not for sale on peptide sites), you'd have to dose below 65MCG. It is unclear if Kisspeptin-10, the widely available analog of Kisspeptin, can cause a similar degeneration. The studies on it are largely IV bolus injections, which do not show testicular degeneration.

    So, my dr. Did not test to see if my primary amenorrhea (no puberty onset) was pituitary or hypothalamus driven. I have low thyroid and low cortisol levels very low FSH, LH, estradiol and progesterone, no GH. I do not menstruate or ovulate, hormone replacement therapy has caused the shedding of the lining of the uterus but no ovulation. I am not currently taking hormone placement therapy. I desire to conceive, I am open to trials of kisspeptin. I am wondering if you have any information that could help me.

    What people don't understand is that vitamin D studies, are purposely designed with extremely low test levels of vitamin D. This is because the US government won't allow high dose studies, like 20,000 IU daily. The FDA has to approve studies, and I have watched doctors say that the government is really strict on making tester follow the rules. Since an average of zero people per year die from vitamin D toxicity, and humans can get 20,000 IU daily of vitamin D3 from the sun, why is the government not allow high dose testing? This tells me they are afraid of something. Maybe the government is trying to keep the beneficial effects of vitamin D from being known. This would make sense. If vitamin D was protective of viruses, then the drug companies would lose billions of dollars. Is it possible the government is protecting their funding sources? Are the drug and medical communities paying off the government to keep a cheap and effective treatment a secret so they can profit off of people's illnesses? Does the government have a track record of hiding the truth to keep certain industries profitable? The evidence strongly suggests this is the case. Why is it that every C19 video of any kind has a notice telling the viewer to get the latest information about it from the CDC? UA-cam wants everybody to blindly just trust the government like a bunch of unquestioning morons without any thought. This is how misinformed people think...

    The fact that causation hasn't been proven with more than 95% certainty isn't the same as there "being no link". I love how linear regression is used to show that the vitamin d correlation is really explained by "real" risk factors such as obesity or ethnicity(which are not independent variables since they correlate with vitamin d levels, and not linear either, and thus cannot be corrected for). To be honest, you could just as well do this the other way around, and then vitamin d would be the real risk factor and the correlation with weight and ethnicity would vanish. There are lies, damn lies, and then there are statistics.

      The plain fact is that Vitamin D levels are below optimal in over 80% of black people in the US, and that rather than notice this obvious correlation with their 2X likelihood of dying from coronavirus, researchers have chosen to keep quiet in favor of profits made on expensive new vaccines. You can't profit from vitamin D capsules, simple as.

    This video is garbage. Spending a few minutes outside and eating an occasional egg salad or bit of salmon will NOT get you anywhere near healthy levels, presumed to be 50ng and above. To reach these levels, you need deliberate sun bathing, or supplements on the order of thousands of IU per day.

  • @geonerd
    @geonerd 3 роки тому

    400 iu per day is a pathetic joke. Ancestral VD levels are generally above 50ng/ml. To achieve this level, you will need to take many thousands of IU per day or sunbathe regularly. (One sun session can generate tens of thousands of IU in under an hour.)

      ... shocking how this topic is treated and in all truth practucal neglected ... I get the impression that this topic is also literally SUPPRESSED ... and it is a long tradition of trying to keep Vitamin D supplementation on a very small level in the public ... the 400 IU is one of those signs (a very dangerous joke)....there are thousands of good studies, which show the big importance and benefit of Vitamin D especially within the Immunsystem ... and it should be told that it is very important to keep a level in the blood of at least 50 to better nearly 100 ng/ml (i keep this level since years and i am never ill since that time - today 62yrs of age) ... not to forget the tremendous importance of MAGNESIUM as ESSENTIAL COFACTOR to give in addition, beside Vit K2 and a bit of Zinc .... my family and I take 10.000 IU per day since many years and about 300-600mg Magnesium daily ... K2 about 150-200 microgram (it is responsible to help to bring and keep the calcium in the bones) .. where are those informations on the English spoken community e.g. on UA-cam.. the are tons of those Informations in German language ... it is well known, that big Pharma doesn't want Vitamin D to be used because it would destroy their market of a lot of products becoming obsolet ....

      I've been supplementing Vit D3 since late 2013. Today most of my family members do the same. Most of us take 10.000 IU daily. Those of us who are obese take up to 25.000 IU daily (prescribed by a doctor who is an expert in the matter). We do blood tests regularly and we are all fine. Not a single case of intoxication or elevated calcium.

    As a patient with Kallmann syndrome this video is of great interest. I have been involved as a patient in two clinical trials using Kisspeptin. Early diagnosis of Kallmann syndrome / CHH can be so important to patients so the use of the Kisspeptin challenge test to distinguish a case of Kallmann syndrome / CHH from standard delay of puberty could be very beneficial to patients. I did not respond to Kisspeptin injections, as would be expected in a Kallmann syndrome patient, even though I responded to GnRH injections.
