I've been a little heavy-handed with my mechanical inoculation and have caused a significant amount of damage to the leaves. Having seen this video I will prepare the next batch with a gentler approach.
Can we refrigerate the prepared serial dilution to be used for tommorow?

I a 13 year old girl doing a science fair experiment. I am inoculating my plants with TMV suspended in milk. My directions suggest that I rinse my leaves after applying TMV. Since I can not rinse the milk from the leaves in my experiment I rinse the abrasion powder before applying the virus. I am wondering if the light tapping that I use to apply the diatomaceous earth will be enough to abrade the leaf before applying TMV dilutions. I can not see signs of abrasion and am worried that my plants will not infect.

Why was the 1ml removed from the last bottle and discarded? The concentration of cells, i.e. cells/ml in that diluent is the same before and after removing the 1ml.

why the dilution becomes 10 times smaller on the plate?
Because the plate contains 9 parts of material and you're adding 1 part of the diluted mixture.
